Expression in donor monocyte derived cells

ABSTRACT: Monocyte differentiate into macrophages that display either pro-inflammatory or tolerogenic profile depending on the inflammatory condition of the intestine. We performed adoptive monocyte transfer to dissect the dynamics of monocyte to macropahges differentiation. Gene expression analysis of donor monocyte-derived cells recovered from non-inflamed and inflamed small intestine of recipient mice at day 1, 2 and 5 after transfer for in-depth investigation of monocyte development in the intestine.

Project description:In this study we showed that Trem1 is one of genes that were differentially regulated in donor monocytes recovered from non-inflamed and inflamed small intestine of recipient mice. We, therefore, aimed at investigating the role of Trem1 on regulating the monocyte adapatation and differentiation in a context-dependent manner. Thus, the gene expression of WT and Trem1-deficient donor monocytes that are recruited to non-inflamed and inflamed small intestine one day after transfer.

Heightened turnover and failed maturation of monocyte-derived macrophages in Chronic Granulomatous Disease

Project description:Loss of NADPH oxidase activity in phagocytes leads to altered cellular responses and exaggerated inflammation in Chronic Granulomatous Disease (CGD). We sought to assess the effects of Nox2 absence on monocyte-derived macrophages (MoMacs) in gp91phox-/y mice during zymosan-induced peritonitis. MoMacs from CGD and wild type (WT) peritonea were lavaged and characterized over time. Though the numbers harvested from both genotypes were virtually identical, there were marked differences in maturation: newly recruited WT MoMacs rapidly enlarged and matured with loss of Ly6C and gain of MHCII, CD206 and CD36, while MoMacs in CGD remained small and were mostly Ly6C+MHCII-. RNAseq analyses showed few intrinsic differences between genotypes in newly recruited MoMacs, but significant differences over time. WT MoMacs demonstrated changes in metabolism, adhesion and reparative functions, while CGD MoMacs remained inflammatory. Labeling with PKH dye demonstrated that while WT MoMacs were mostly recruited within the first 24 hours and remained in the peritoneum while maturing and enlarging, CGD monocytes continued to stream into the peritoneum for days with many migrating to the diaphragm where they were found in fibrin(ogen) clots surrounding clusters of neutrophils in what appeared to be nascent granulomata. Importantly, these observations appeared to be entirely driven by the milieu: adoptive transfer of CGD MoMacs into inflamed peritonea of WT mice resulted in immunophenotypic maturation and behavior, and conversely, altered maturation/behavior of WT MoMacs was seen after adoptive transfer into inflamed peritonea of CGD mice. These data demonstrate heightened recruitment and fundamental failure of MoMac maturation in CGD.

GATA3 induces the pathogenicity of Th17 cells via regulation of GM-CSF expression

Project description:To investigate the effects of a late deletion of Gata3 on CD4 T cell gene expression profiles in experimental autoimmune encephalomyelitis, we performed a RNA-Seq analysis of Gata3-sufficient (i.e., Cd45.1/Cd45.2 or vehicle treated Cd45.2/Cd45.2 Cre-ERT2 Gata3 fl/fl) and Gata3-deficient (i.e., tamoxifen-treated Cd45.2/Cd45.2 Cre-ERT2 Gata3 Fl/Fl) CNS-infiltrating CD4+ effector T cells from mixed congenic co-transfer recipient mice. We performed a gene expression profiling analysis using data obtained from RNA-seq from 2 mixed congenic adoptive transfer receipient vehicle-treated mice and 2 mixed congenic adoptive transfer receipient tamoxifen-treated mice for a total of 8 biological samples.

Transcriptomes of monocyte-derives DCs stimulated with various compounds

Project description:Little is known about the early transcriptional events in innate immune signaling in immature and tolerogenic monocyte-derived dendritic cells (DCs), the professional antigen-presenting cells of our immune system. TLR ligands usually induce a proinflammatory transcriptional response, whereas IL10 and/or dexamethasone induce a more tolerogenic phenotype. We used Affymetrix microarrays to obtain detailed information underlying pro- and anti-inflammatory transcriptional responsesand transcriptional networks in DCs A pilot experiment was performed in which monocyte-derived DCs were either treated with TLR4 ligand LPS, or IL10 and dexamethason. Furthermore, IL10/dexamethason treated cells were also stimulated with LPS for an additional 6 hr. All samples were then subjected to global gene expression analysis using Affymetrix technology.

TLR2 dimerization blockade allows generation of homeostatic intestinal macrophages under acute colitis challenge

Project description:Recruited blood monocytes contribute to the establishment, perpetuation and resolution of tissue inflammation. Specifically, in the inflamed intestine, monocyte ablation was shown to ameliorate colitis scores in preclinical animal models. However, the majority of intestinal macrophages that seed the healthy gut are also monocyte-derived. Monocyte ablation aimed to curb inflammation would therefore likely interfere with intestinal homeostasis. Here, we used a TLR2 trans-membrane peptide which blocks TLR2 dimerization that is critical for TLR2 /1 and TLR2 / 6 heterodimer signaling to blunt inflammation in a murine colitis model. We show that while the TLR2 peptide treatment ameliorated colitis, it allowed recruited monocytes to give rise to macrophages that lack the detrimental pro-inflammatory gene signature and reduced potentially damaging neutrophils infiltrates. Finally, we demonstrate TLR blocking activity of the peptide on in vitro cultured human monocyte-derived macrophages. Collectively, we provide a significantly improved anti-inflammatory TLR2 peptide and critical insights in its mechanism of action towards future potential use in the clinic.

Carvacrol treated Dentritic Cells

Project description:Tolerogenic dendritic cells (DCs) induce regulatory T-cells and dampen pathogenic T-cell responses. Hereby, they have gained interest as a therapeutic target in the combat against autoimmune diseases. In this study we investigated whether tolerogenic DCs are induced by the phytonutrient carvacrol, a molecule with known anti-inflammatory properties. In this study, bone marrow derived DCs were treated with carvacrol in combination with thermal stress. Gene expression profiles were obtained by microarray analysis to test for an induced tolerogenic phenotype. To investigate the tolerogenic properties of treated DCs in vivo, T-cell anergy or the induction of a regulatory T-cell phenotype was studied in antigen specific T-cells. Finally, treated DCs were tested by transfer into an experimental arthritis model.

BMI1 Maintains the Regulatory T cell Epigenomic Landscape to Prevent Inflammatory Bowel Disease (Microarray)

Project description:FOXP3+ Treg cells are expanded within the inflamed intestine of human Crohn’s disease, yet FOXP3-mediated gene repression within these cells is lost. The Polycomb repressive complexes play a role in FOXP3 target gene regulation, but deeper mechanistic insight is incomplete. We have now specifically identified the Polycomb Repressive Complex 1 (PRC1) family member, BMI1 in the regulation of a pro-inflammatory enhancer network in both human and murine Tregs. Using human Tregs and lamina propria T cells, we inferred PRC1 to regulate Crohn’s associated gene networks through assays of chromatin accessibility. Conditional deletion of BMI1 in murine FOXP3+ cells led to systemic inflammation. BMI1-deficient Tregs beared a TH1/TH17-like phenotype as assessed by assays of genome wide transcription, chromatin accessibility and proteomic techniques. Finally, BMI1 mutant FOXP3+ cells did not suppress colitis in the adoptive transfer model of human inflammatory bowel disease. We propose that BMI1 plays an important role in enforcing Treg identity in vitro and in vivo. Loss of Treg identity via genetic or transient BMI1 depletion perturbs the epigenome and converts Tregs into Th1/Th17-like proinflammatory cells, a transition relevant to human Crohn’s disease associated CD4+ T cells.
