ABSTRACT: Obesity and type 2 diabetes (T2D) can be associated with altered secretion of enterohormones in condition that remains to be understood in depth. Here, we aimed to decipher the mechanisms by which a major enterohormone GLP-1, is decreased in human obese patients according to their diabetic status.

GLP-1 directed NMDA receptor antagonism for obesity treatment

Project description:The N-methyl-D-aspartate (NMDA) receptor is a glutamate-activated cation channel critical to many processes in the brain. Genome-wide association studies (GWAS) suggest that glutamatergic neurotransmission and NMDA receptor-mediated synaptic plasticity is important for body weight homeostasis1. Here, we report the engineering and preclinical development of a first-in-class bimodal molecule that integrates NMDA receptor antagonism with glucagon-like peptide-1 (GLP-1) receptor agonism to effectively reverse obesity, hyperglycemia, and dyslipidemia in rodent models of metabolic disease. We demonstrate that GLP-1-directed delivery of the NMDA receptor antagonist MK-801 affects NMDA receptor-mediated synaptic plasticity in the hypothalamus. Importantly, peptide-targeting of MK-801 specifically to GLP-1 receptor-expressing brain regions circumvent adverse physiological and behavioral effects associated with MK-801 monotherapy. In sum, our approach demonstrates the feasibility of cell specific ionotropic receptor-modulation via peptide targeting and highlights the therapeutic potential of unimolecular mixed GLP-1 receptor agonism and NMDA receptor antagonism for obesity treatment.

Project description:The peroxisome proliferator-activated receptors alpha and gamma (PPAR?/?) play important roles in the regulation of energy, lipid and glucose metabolism. Drugs targeting these proteins such as Fibrates and TZDs have both shown beneficial cardiovascular effects in clinical trials, which together with their complementary action on glucose and lipid metabolism spurred the development of PPAR?/? dual-agonists (Glitazars) for the treatment of type-2 diabetes (T2D) and dyslipidemia. While effectively improving glucose and lipid metabolism in clinical trials, the development of many Glitazars was terminated due to unfavorable effects on the cardiovascular and/or renal system. Here we evaluate a single molecule conjugate of the PPAR?/? dual-agonist Tesaglitazar covalently attached to the incretin hormone glucagon-like peptide-1 receptor agonist (GLP-1RA). We hypothesized that GLP-1-mediated delivery of Tesaglitazar as well as synergism between the two agents would lead to overal beneficial effects and decrease the TZD risk profile.

GLP-1 signaling suppresses menin's transcriptional block by phosphorylation in beta cells

Project description:We find that GLP-1 signaling-activated PKA directly phosphorylates menin at serine 487 residue, relieving menin mediated suppression of insulin expression in beta cells and islets of diabetic GK rat. Mechanistically, GLP-1-induced PKA activation leads to menin Ser487 phosphorylation, and phosphorylated menin gains increased binding affinity to cytoskeleton proteins such as beta actin and myosin. Sequestration of Ser487 phosphorylated menin from the Ins1 gene promoter leads to reduced binding of menin-associating repressive epigenetic regulators such as SUV39H1 and HDAC1 at the locus, resulting in increased Ins1 transcription. Our results have linked GLP-1 signaling to physiologically suppression of menin function in repressing insulin expression, and uncovered a potential step to modulate menin phosphorylation to improve T2D therapy.
