HSD17B14 induction in breast cancer cell lines

Project description:The 17beta-hydroxy steroid dehydrogenase family is involved in the fine tuning of conversion of different substances such as steroids, retenoids and fatty acids. The expression of HSD17B14 has been correlated to better prognosis in breast cancer. The function of HSD17B14 is still not known, but it may convert estradiol to estrone. We transfected breast cancer cell lines with a HSD17B14-containing plasmid and compared with mock-transfected samples. The influence on global gene expression was examined with microarrays. The breast cancer cell lines SKBR3 and ZR75-1 were transfected with a HSD17B14-containing plasmid or a plasmid without insertion as control. This was performed in duplicates. The cells were harvested after 48 hours, and RNA was extracted and hybridized on Affymetrix microarrays.

We examined any genetically driven sex dimorphisms in human pluripotent stem cells focusing on Y chromosome contribution. Project description:This study was designed to define erythropoietin (EPO) regulated genes in murine bone marrow erythroid progenitor cells at two stages of development, designated E1, and E2. E1 cells correspond to CFUe- like progenitors, while E2 cells are proerythroblasts. 14 samples were analyzed. 8 samples belong to the E1 stage and the remaning 6 samples correspond to the E2 stage. In the E1 stage, there are 4 controls and 4 treatment samples. The 4 samples in each group are biological replicates. Likewise in the E2 stage, there are 3 control and 3 treatment samples and the 3 samples in each group are biological replicates.

Gene expression of clonal subpopulations derived from the 4T1 murine mammary carcinoma cell line

Project description:Breast tumors often exhibit intratumoral heterogeneity. We hypothesized that phenotypically distinct clonal subpopulations in heterogeneous tumors may affect overall tumor morphology and cancer cell invasion. In order to study heterogeneous tumors, we isolated individual cells from a parental 4T1 murine mammary carcinoma cell line and generated four clonal subpopulations (E1, E2, A, and M). To characterize these subpopulations, we examined gene expression by performing bulk RNA-sequencing.
