Small RNA sequencing of LA-N-2 and LAN-5 cells exposed to ciliary neurotrophic factor for 30/60 min and 2/4 days

ABSTRACT: To examine the impact of gp130-mediated, pro-cholinergic neurokine intervention on the small RNA systems in female and male human neuronal cells, we exposed cultured LA-N-2 cells (female) to 100 ng/ml ciliary neurotrophic factor (CNTF), and LAN-5 cells (male) to 10 ng/ml CNTF. RNA sequencing of time points at 30/60 minutes and 2/4 days of CNTF exposure shows a significant response and an “immediate early”/”long term” dichotomy in differentially expressed miRNAs.

Impacts of ciliary neurotrophic factor on the retinal transcriptome in a mouse model of photoreceptor degeneration

Project description:Ciliary neurotrophic factor (CNTF) has been tested in clinical trials for human retinal degeneration due to its potent neuroprotective effects in various animal models. To decipher CNTF-triggered molecular events in the degenerating retina, high-throughput RNA sequencing analyses were performed using the Rds/Prph2 (P216L) transgenic mouse as a preclinical model for retinitis pigmentosa. Retinal transcriptome data was obtained for untreated wild type retinas, Rds retinas treated with a lentivirus expressing CNTF from postnatal day 25 to 35, and Rds retinas similarly treated with a control lentivirus expressing GFP. Comparisons of transcriptome data detect significant changes between wild type and Rds retinas. In addition, RNA-seq data reveal elevation of transcripts in the innate immune system and growth factor signaling, as well as altered neuronal transmission and metabolism in CNTF-treated Rds retinas. These results demonstrate the influence of exogenous CNTF on the retinal transcriptome landscape and illuminate likely CNTF impacts in degenerating human retinas.

Project description:Our data suggest that CNTF remodels the transcription profile of Muller (glial) cells leading to induction of networks associated with transcription, cell cycle regulation and inflammatory response. CNTF also appears to function as an inducer of gliosis in the retina. These studies provide new insights into the biological functions of cytokines in the retina. Project description:<p>To learn about the etiology of lung cancer among never-smoking women in Asia, we formed the Female Lung Cancer Consortium in Asia (FLCCA), which includes studies of lung cancer in Eastern Asia and conducted a GWAS. We analyzed a total of 5510 lung cancer cases and 4544 controls and identified 3 novel loci (Lan et al., 2012). Of these study subjects, 4922 lung cancer cases and 3959 controls are available for posting.</p> <p>Lan et al.., Genome-wide association analysis identifies new lung cancer susceptibility loci in never-smoking women in Asia.
