Measuring the transcriptomic response to targeting kinases correlated with estrogen receptor activity in Breast Cancer

ABSTRACT: We undertook a phosophotranscriptomic network analysis of published patient tumour RNA-seq and phosphoproteomic data to identify kinases that correltated with Estrogen Receptor activity in Breast Cancer. The most sucessful canidates were selected for targeting by siRNA and the effects monitored by RNA-seq. Comparison the transcriptional changes in the mutation of VZV circVLTs

Project description:Sequence analyses shown that most exons of VZV circVLTs were overlapped with ORFs, except for exon5, which was an ideal mutation position. Using a galK positive/counter selection VZV bacteria artificial chromosome (BAC) system, we replaced upstream (104,765-104,785 Mutation2, pOKA-M2) or downstream (104,785-104,805, Mutation1, pOKA-M1) of circVLTs BSJ exon5 with 20 bp random sequences in VZV genomic DNA level. Discovery of Varicella zoster virus circular RNA with nanopore sequencing

Project description:Varicella-zoster virus (VZV), an alphaherpesvirus, causes chickenpox (varicella) in young children with an annual minimum of 140 million new cases and herpes zoster in senior, a painful and debilitating disease with 3-5‰ incidence. A complex structural transcriptome of VZV, which numerous novel transcripts, transcript isoforms, and unknown splice events are found during cell infection. Circular RNA (circRNA), a newly important component of the transcriptome, is increasing discoveries of circRNA function in mammalian cells. However, VZV encoded circRNA remains unexplored.
