Project description:Despite the importance of egg development to the female life cycle in Drosophila, global patterns of gene expression have not been examined in detail primarily due to the difficulty of synchronizing developmental stages. Entry into vitellogenesis is however an key stage of oogenesis, and by delaying entry past this control point, we have been able to investigate some of the transcriptional dynamics apparent before and after early egg formation over a 72 hour period. A short-day photoperiod (10:14 light:dark photoperiod) and moderate temperatures (11°C) are required for the induction of ovarian diapause. For our microarray experiments, 0-4 hours post-eclosion females were immediately introduced to the restrictive environmental conditions. Females were subsequently collected for RNA extraction after 5-7 days, or were alternatively re-introduced to normal environmental conditions to continue egg development. Time-points were collected before and after the release of reproductive arrest at 24 hour intervals and hybridized in a balanced loop design, including dye-swaps. Eight biological replicates were subsequently performed for each treatment, including 3 day old male and female controls maintained under normal environmental conditions.

Project description:During Drosophila melanogaster oogenesis, the JAK/STAT and EGFR pathways are both required to specify a population of somatic epithelial cells called the posterior follicle cells (PFCs). The PFCs are important because they generate a signal at mid-oogenesis that is required to establish the embryonic axes. To identify novel PFC-expressed transcripts, egg chambers containing ectopic PFCs were generated and microarrays were then used to identify upregulated transcripts. To generate ectopic PFCs, GAL80TS / CyO ; UAS-Upd, UAS-λtorpedo/TM6B flies were crossed to GR1-GAL4 flies to obtain the genotype GAL80TS/+ ; GR1-GAL4/UAS-Unpaired, UAS-λtorpedo. Sibling females of genotype GAL80TS/+ ; GR1-GAL4/TM6B were dissected as controls.

Project description:Ribosomes are produced in large quantities during oogenesis and stored in the egg. However, the egg and early embryo are translationally inactive. How translation is activated during embryogenesis is poorly understood. Using mass-spectrometry and cryo-EM analyses of ribosomes isolated from zebrafish and Xenopus eggs and embryos, we find that ribosomes transition from a dormant to an active state during the first hours of embryogenesis. Dormant ribosomes are associated with four factors that form two modules: a Habp4-eEF2 module that stabilizes ribosome levels and a Dap1b/Dapl1-eIF-5a module that represses translation. Dap/Dap1b is a newly discovered translational inhibitor that stably inserts into the polypeptide exit tunnel. Thus, a developmentally programmed, unique ribosome state plays a key role in ribosome storage and translational repression in the egg.

Project description:We used a transcriptomic approach based on the comparison of the expression between the liver of sexually mature hens versus pre-laying pullets to better appreciate which hepatic proteases and antiproteases are specifically expressed in relation to vitellogenesis. Using a 20K chicken oligoarray corresponding to 12 595 different chicken genes, a total of 582 genes were shown to be over-expressed in the liver at sexual maturity of hens (1.2 to 67 fold- difference). Most of the top ten over-expressed genes are known components of egg yolk or of the perivitelline membrane. The combination of different bioinformatic tools reveals 12 proteases and 3 antiproteases amongst the over-expressed genes, including many predicted proteins with yet unknown functions.

This Ca2+ rise sets off a series of downstream events, including the completion of meiosis and the dynamic remodeling of the oocyte transcriptome and proteome, which prepare the oocyte to undertake embryogenesis. Calcineurin is a highly conserved phosphatase that is activated directly by Ca2+ upon egg activation and that is required for the resumption of meiosis in Xenopus, ascidians and Drosophila. The molecular mechanisms by which calcineurin transduces the calcium signal to regulate meiosis and other downstream events are still unclear. In this study, we investigate the regulatory role of calcineurin during egg activation in Drosophila melanogaster. Using mass spectrometry, we quantify the phosphoproteomic and proteomic changes that occur during egg activation, and we examine how these events are affected when calcineurin function is perturbed in female germ cells. Our results show that calcineurin regulates hundreds of phosphosites and also influences the abundance of numerous proteins during egg activation. We find calcineurin-dependent changes in cell cycle regulators including Fzy, Greatwall (Gwl) and Endosulfine (Endos), protein translation modulators including PNG, NAT, eIF4G and eIF4B, and important components of signaling pathways including GSK3β and Akt1. Project description:Hytrosaviridae family members replicate in the salivary glands (SGs) of their adult dipteran hosts and are transmitted to uninfected hosts via saliva during feeding. Despite inducing similar gross symptoms (SG hypertrophy; SGH), hytrosaviruses (SGHVs) have distinct pathobiologies, including sex-ratio distortions in tsetse flies and refusal of infected housefly females to copulate. Via unknown mechanism(s), SGHV replication in other tissues results in reduced fecundity in tsetse flies and total shutdown of vitellogenesis and sterility in housefly females. We hypothesized that vitellogenesis shutdown is caused by virus-induced modulation of hormonal titers. Here, we used RNA-Seq to investigate virus-induced modulation of host genes/pathways in healthy and virus-infected houseflies, and we validated expression of the most significantly modulated genes (n=23) by RT-qPCR. We also evaluated the levels and activities of hemolymph antimicrobial peptides (AMPs), levels of endogenous sesquiterpenoids, and impacts of exogenous hormones on ovarian development in viremic females. Of the 973 housefly unigenes that were significantly modulated (padj ≤ 0.01; log2FC ≤ or ≥ 2.0 ), 446 and 527 genes were downregulated and upregulated, respectively. While the most downregulated genes were related to reproduction (embryogenesis/oogenesis), the repertoire of upregulated genes was overrepresented by genes related to non-self recognition, ubiquitin-protease system, cytoskeletal traffic, cellular proliferation, development and movement, and snRNA processing. Overall, MdSGHV induced upregulation of components of the siRNA, innate antimicrobial immune, and autophagy pathways. MdSGHV reduced hemolymph sesquiterpenoids and completely shut down egg development in viremic females. However, the hormonal rescue of vitellogenesis did not result in egg production. The mechanism underlying MdSGHV-induced sterility has yet to be resolved.
