Microglia and infiltrating monocytes gene expression profiling during EAE progress

ABSTRACT: The project was designed to figure out the role of microglia in brain inflammation (EAE model). The first step was to find genes/pathways that are upregulated in the microglia during EAE progress but absent in the infiltrating monocytes. These genes/pathways may provide clues for studying the unique function of microglia in the brain inflammation that is disctinct from the infiltrating monocytes.

Project description:The role of microglia and infiltrating monocytes in experimental autoimmune encephalomyelitis (EAE) pathogenesis has been controversial. To gain insight into their respective roles, we developed a method for differentiating between microglia and infiltrating monocytes in the CNS using CD44. We used this system to monitor changes in cell number, activation status, and gene expression by RNA sequencing (RNA-Seq) over the course of disease. This in vivo characterization and RNA-Seq dataset improves our understanding of myeloid cell biology in the brain under inflammatory conditions and may lead to strategies to identify therapies for inflammatory pathways active in neuroinflammatory diseases. Pooled samples from 10 mice were analyzed for both microglia and monocytes at distinct timepoints post EAE inducation. Peritoneal macrophages were isolated and analyzed for five samples, and also in a pool.

Pharmacologic inhibition of EphB3 receptor kinase ameliorates EAE

Project description:To evaluate the potential of EphB3 as a therapeutic target during CNS inflammation, we induced EAE in B6 WT mice and starting at the peak of the disease we initiated treatment with A38 administered. A38 administration ameliorated EAE, and the transcriptional analysis of the astrocytes and microglia revealed the decreased expression of genes associated to inflammation and neurodegeneration.

Autonomous TNF is critical for monocyte survival in steady state and inflammation in vivo

Project description:Monocytes are circulating mononuclear phagocytes, poised to extravasate to sites of inflammation and differentiate into monocyte-derived macrophages and dendritic cells. Tumor necrosis factor (TNF) and its receptors are upregulated during monopoiesis and expressed by circulating monocytes, as well as effector monocytes infiltrating certain sites of inflammation, such the spinal cord during experimental autoimmune encephalomyelitis (EAE). Using competitive in vitro and in vivo assays, we show here that monocytes deficient for TNF or TNF receptors are outcompeted by their wild-type counterpart. Moreover, monocyte autonomous TNF is critical for the function of these cells, as TNF ablation in monocytes / macrophages, but not in microglia delayed the onset of EAE in challenged animals and was associated with reduced acute spinal cord infiltration of Ly6Chi effector monocytes. Collectively, our data reveal a previously unappreciated critical cell-autonomous role of TNF on monocytes for their survival, maintenance and function.

Microarray of brain microglia from GM-CSF-expressing B cell conditional knock-out mice at EAE peak stage

Project description:We found a kind of GM-CSF producing B cells (BGM) is positively correlated with the pathogenesis of EAE. BGM cell conditional knock-out (CKO) mice showed significant resistance to EAE. To investigate the relationship between BGM cells and microglia and comprehensively evaluate the status of microglia, we obtained microglia from the brain of CKO and control mice at EAE peak stage, extracted microglial RNA, and performed microarray analysis.

Treatment with nanoliposomes loaded with ITE and MOG(35-55) ameliorates EAE

Project description:To evaluate the therapeutic potential of NLP(ITE+MOG) in a mouse model of secondary progressive multiple sclerosis, we induced EAE in NOD mice and initiated weekly treatment with nanoliposomes beginning at 35 days post immunization during the chronic phase of disease. NLP(ITE+MOG) administration ameliorated EAE, and the transcriptional analysis of CNS-resident astrocytes, microglia and monocytes revealed decreased expression of genes associated with pro-inflammatory responses and neurodegeneration.

DNA methylome and transcriptome sequencing to define pathways involved in the activation of microglia

Project description:Microglia cells are macrophages which reside in the brain, and become activated during the inflammatory processes involved in neurodegenerative and autoimmune disorders. An understanding of the molecular processes involved in this activation process could lead to the development of new neuroprotective therapies. To reveal molecular mechanisms involved in microglia activation, we performed RNA-seq and MBD-isolated genome sequencing (MiGS or MBD-seq) on both naïve and peak microglia derived from the mouse experimental autoimmune encephalomyelitis (EAE) model. The EAE model reflects some of the histological and immunological features of multiple sclerosis (MS) in humans. Mice genetically engineered to label Ccr2 with red fluorescent protein (rfp) and Cx3cr1 with green fluorescent protein (gfp) (“red-green mice” as described in Saederup et al., 2010 and Mizutani et al., 2012) were employed and microglia (positive for gfp) were separated from infiltrating monocytes using flow cytometry.

Cognate Microglia - T cell interactions shape the functional regulatory T cell pool in EAE pathology

Project description:Microglia, the parenchymal brain macrophages of the central nervous system (CNS), have emerged as critical players in brain development and homeostasis. Immune functions of these cells, however, remain less well defined. We investigated contributions of microglia in a relapsing remitting (RR) multiple sclerosis paradigm, experimental autoimmune encephalitis (RR-EAE) in C57BL/6 / SJL F1 mice. Fate mapping-assisted translatome profiling during the RR disease course revealed the potential of microglia to interact with T cells through antigen presentation, co-stimulation, and co-inhibition. Abundant microglia - T cell aggregates, as observed by histology and flow cytometry, supported the notion of functional interactions of microglia and T cells during remission, with a bias towards T regulatory cells. Finally, microglia_x0002_restricted Ifng receptor and MHC mutagenesis significantly affected the functionality of the regulatory T cell compartment in the diseased CNS and remission. Collectively, our data establish critical non-redundant cognate and cytokine-mediated interactions of microglia with CD4+ T cells during autoimmune neuro-inflammation.
