Expression data from murine bone marrow-resident plasma cells and spleen mature follicular B cells

Project description:CD138+ B220- plasma cells were sorted from bone marrow and B220+ CD23+ mature follicular B cells were sorted from the spleens. Plasma cells were sorted from C57BL/6 mice 7 days after boosting with antigen, with mice first primed with an i.p. injection of KLH/IFA followed by boost at day 21 with KLH/PBS i.p. Mature B cells were sorted from antigen-naïve C57BL/6 mice. We compared expression profiles of plasma cells and mature B cells to identify differentially expressed transcripts. Seven days after boosting with KLH, plasma cells were enriched by CD138-PE/anti-PE magnetic bead enrichment and then sorted to purity. Affymetrix microarrays were performed on 50,000-70,000 plasma cells and 2,000,000-3,000,000 naïve B cells.

Project description:Plasma cell gene expression is driven both by isotype and tissue location. In this series we examine gene expression of bone marrow IgA, IgM and IgG plasma cells as well as IgA plasma cells from small intestine lamina propria. To validate tissue specific gene expression we also include gene expression from lamina propria IgA-/- plasma cells. All plasma cell samples are from Blimp1+/GFP reporter animals and splenic follicular and marginal zone B cell gene expression have been added as reference populations.

CTCF binding in B cells and Plasmablasts

Project description:The mammalian CCCTC-binding factor (CTCF) regulates gene expression through the formation of higher order chromatin structures. Recent evidence has implicated a role for CTCF in regulating gene expression in the human MHCII locus. To investigate the role of CTCF in murine MHCII gene expression we mapped CTCF binding sites in B cells (MHCII+ cells) and Plasmablasts which are differentiated B cells that have silenced MHCII gene expression. These observations lead to the identification of differential CTCF binding during differentiation in these cell types and suggest mechanims of MHCII gene regulation. Project description:Observations that ongoing mTORC1 activity drives plasma cell poise in MZ B cells we rationed that enforcing mTORC1 activity on mature follicular B cells by inducible B cell specific Tsc1 deletion would confer plasma cell differention poise on follicular B cells. We therefore performed RNA-seq on follicular and marginal zone B cells from Tsc1F.hCD20-TamCre mice after 4 weeks tamoxifen treatment to examine transcriptional markers of plasma cell differentiation poise.

Project description:Plasma cell differentiation is thought to be tied to cell division. We observe that marginal zone B cells are able to differentiate to plasma cells without completing a single cell division. In order to characterize the transcriptome of plasma cells derrived from un-divided MZ B cells we performed in vitro plasma cell differentiation cultures in the presence of the CDK4/6 inhibitor PD0332991 or rapamycin using Blimp1-GFP reporter follicular and MZ B cells. We report here that these resulting plasma cells from undivided MZ B cells exhibit normal plasma cell gene expression and function in the absence of mitosis.
