Expression profiling of isogenic A549 cell lines with and without ELF3 knock-down

Project description:Gene function in cancer is often cell type-specific. The epithelial cell-specific transcription factor ELF3 is a documented tumour suppressor in many epithelial tumours yet displays oncogenic properties in others. Here, we show that ELF3 is an oncogene in the adenocarcinoma subtype of lung cancer (LUAD), providing genetic, functional, and clinical evidence of subtype specificity. We discover a region of focal amplification at chromosome 1q32.1 encompassing the ELF3 locus in LUAD which is absent in the squamous subtype. Gene dosage and promoter hypomethylation affect the locus in up to 80% of LUAD analysed. ELF3 expression was required for tumour growth and a pan-cancer expression network analysis supports its subtype and tissue specificity. We further show that ELF3 displays strong prognostic value in LUAD but not LUSC. We conclude that, contrary to many other tumours of epithelial origin, ELF3 is an oncogene and putative therapeutic target in LUAD.

Differentially-expressed-transcriptome study of Non-Small Cell Lung Carcinoma

Project description:We systematically profiled the genome-wide alternative splicing events in Lung Adenocarcinoma (LUAD) and Lung Squomous Cell Carcinoma (LUSC) against Lung Control through Human Transcriptome Array2.0. Non-invasive Stage IIIA non-small cell lung cancer (NSCLC) is heterogeneous in nature which makes it difficult to predict, diagnose and prognose owing to lower 5-year survival rate and 75-85% brain or bone metastasis. Hence, we hypothesized to develop transcript-based signature to categorize Stage IIIA-NSCLC-LUAD and LUSC, as well as identify markers which could indicate towards prognosis of disease. We were able to molecularly-categorize LUAD- and LUSC-tissue more precisely through HTA array2.0.

FCN3 functions as a tumor suppressor of lung adenocarcinoma through induction of endoplasmic reticulum stress

Project description:In this study, we report a novel function of FCN3 (Ficolin 3), a secreted lectin capable of activating the complement pathway, as a tumor suppressor of lung adenocarcinoma (LUAD). First, the expression of FCN3 was strongly down-regulated in cancer tissues compared to matched normal lung tissues, and down-regulation of FCN3 was shown to be significantly correlated with increased mortality among LUAD patients. Interestingly, while ectopic expression of FCN3 led to cell cycle arrest and apoptosis in A549 and H23 cells derived from LUAD, the secreted form of the protein had no effect on the cells. Rather, we found evidence indicating that activation of the unfolded protein response is induced from ectopic expression of FCN3 which lead to endoplasmic reticulum (ER) stress. Consistently, inhibition of ER stress response led enhanced survival of the LUAD cells. Collectively, our data indicate that FCN3 is a tumor suppressor gene functioning through induction of ER stress.
