Transcriptomic analysis of the soleus muscle extracted from Dnm2 K562E point-mutant heterozygous mice

ABSTRACT: RNA sequencing was performed to evaluate the impact of Dnm2 K562E mutation in the soleus muscle. The assessment of the dataset reveals a large number of differentially regulated transcripts between animals of both genotypes. This is consistent with a dominant phenotype caused by the Dnm2 K562E point mutation on the muscle.

Project description:To uncover novel genes and molecular pathways important for skeletal muscle remodeling and regeneration, we used a mouse hindlimb unloading and reloading protocol. We then performed analysis of global gene expression by RNA-sequencing (RNA-seq), looking at the transition from muscle unloading to reloading (GSE237099). This study focuses on the microprotein Mustn1 (Musculoskeletal embryonic nuclear protein 1, also known as Mustang), which consists of 82 amino acids. We performed RNA-sequencing of soleus muscle from 8-week-old mice carrying one allele (heterozygous) of Mustn1 tm1a (KO-first) and wildtype littermates.

ALCAR supplementation to unloaded soleus muscle

Project description:The effect of acetyl-L-carnitine (ALCAR) supplementation to hindlimb suspended rats has been analyzed in order to test whether this would promote the differentiation towards an oxidative phenotype of muscle fibers in soleus muscle and prevent the transformation of this slow tonic muscle towards a glycolytic, low-oxidative phenotype due to unloading condition. 3 month-old rats were daily supplemented with ALCAR concomitantly with hindlimb suspension. Validated microarray analysis for a broad range of muscle-relevant processes delineated an ALCAR-induced adjustment of the expression profile. In particular, transcript elevations of factors underlying oxidative metabolism in mitochondria and protein turnover by ribosomal and proteasomal components as well as an adaptation of voltage-gated ion channel expression were identified as an ALCAR response in soleus muscle.
