A novel response to microtubule perturbation in meiosis

Project description:Cells were grown to saturation in YPD (YEP + 2% glucose) for 24 hours, diluted into YPA (YEP + 2% potassium acetate) at OD600= 0.3 and grown over night at 30C. Cells were washed with sterilized water the next day and re-suspended in SPII medium (0.3% potassium acetate, pH = 7.0) at OD600= 1.9 to induce sporulation. Cells were sporulated at room temperature or 30C as indicated. Sporulation medium containing benomyl was always prepared freshly on the day of the experiment following the directions in {Shonn, 2000 #90}. Briefly, DMSO (dimethyl sulfoxide, Sigma-Aldrich) or benomyl [Methyl 1-(butylcarbamoyl)-2-benzimidazolecarbamate, Sigma-Aldrich; 30 mg/ml stock in DMSO] was dissolved in near-boiling SPII medium to avoid precipitation. The medium was then allowed to slowly cool to 30C or room temperature. At the time of drug treatment, cells were filtered and immediately re-suspended in the medium containing benomyl or DMSO. Expression data from wild type and ttk^twk mutant whole ovaries

Project description:Epithelial tube morphogenesis requires precise orchestration of cell signaling, shape, migration, and adhesion. Follicle cells in the Drosophila ovary form epithelial tubes, the lumens of which act as molds for the eggshell respiratory filaments, or dorsal appendages (DAs). The Tramtrack69 (TTK69) transcription factor controls DA lumen volume and shape by promoting tube expansion; the tramtrack mutation twin peaks (ttk^twk) reduces TTK69 levels late in oogenesis, inhibiting DA tube expansion. We used microarrays to identify differentially expressed genes in the ttk^twk mutant that are downstream of TTK69 and that play a functional role in DA tube expansion. Ovaries were dissected from female flies at 36 hours of age, to maximize the proportion of developing egg chambers undergoing DA tube expansion. Flies were raised at a constant temperature of 25C in the presence of wet yeast, to maximize the rate of egg chamber production.

Project description:We sequenced mRNA from whole flies of females and male of Drosophila melanogaster, D. simulans, D. yakuba, D. ananassae, D. pseudobscura, D. mojavensis, and D. virilis as part of the modENCODE project to validate novel gene models found in D. melanogaster, identify genes differentially expressed between the sexes and sex-specific alternative splicing events, and examine the evolution of gene expression. Comparison of expresssion profiles in female and male whole flies. These are the same samples used to generate the array data found in GSE6640.
