ABSTRACT: In order to explore the underlying molecular mechanisms of the impaired olfactory behaviors demonstrated in mice with knockdown of miR-200b/a in the MOE, we dissected the MOE tissues of the miR-200b/a knockdown mice and their NC controls for RNA transcriptome analysis.

Project description:Metastasis associated 1 family, member 2 (MTA2) gene is classified to metastasis associated gene family. We have previously reported that MTA2 gene was overexpressed in gastric cancer tissues, correlating with tumor invasion, lymph node metastasis, and advanced TNM stage. MTA2 knockdown significantly inhibited gastric cancer cell invasion and metastasis. Yet, its molecular mechanisms are still unclear. The aim of this study is to investigate the molecular mechanisms of MTA2 in regulating malignant behaviors of gastric cancer. This experiment captures the expression data between BGC-823/NC and BGC-823/MTA2, SGC-7901/NC and SGC-7901/shMTA2 cells using Whole human genome microarray 4×44K (Design ID: 014850, Agilent technologies).

miR-200b is associated with cisplatin sensitivity in bladder cancer cells

Project description:We aimed to clarify the role of miR-200b in cisplatin (CDDP) sensitivity in bladder cancer (BCa). CDDP resistant T24 cells (T24RC) were transfected with a miR mimic negative control (NC) or a miR-200b mimic, after which cells were treated with or without CDDP. We found that ectopic miR-200b expression re-sensitized the T24RC cells to CDDP. Gene expression microarray analysis revealed that the combination of miR-200b and CDDP affected genes involved in CDDP sensitivity and cytotoxicity.

Data for miR-429, miR-205, miR-200b, and miR-141 were each compared independently to the same negative control data contained in the raw .CEL files nc-miR-1-1.CEL, nc-miR-1-2.CEL and nc-miR-1-3.CEL. For each miRNA, the negative control data were re-normalized together with data only from that specific miRNA. The experiment for M12 was performed at a later date and an independent set of negative control data was collected at that time. Therefore, only M12 data was compared to the negative control data contained in the raw .CEL files nc-miR-2-1.CEL, nc-miR-2-2.CEL, and nc-miR-2-3.CEL.

Project description:Ribosomal protein (RP) L23 is a negative regulator of cell apoptosis. RPL23 overexpression is associated with abnormal apoptotic resistance in CD34+ cells derived from patients with higher-risk myelodysplastic syndrome (MDS). However, the mechanism underlying RPL23-induced apoptotic resistance in higher-risk MDS patients is poorly understood. Gene microarray analysis between RPL23-knockdown (RPL23-KD) and matched control cells (RPL23-NC) was performed to detail global gene expression profiles and to identify differentially expressed genes and potentially involved pathways associated with RPL23 knockdown.

Transcriptome of the MOE in different mouse strains

Project description:High-throughput RNA sequencing of the main olfactory epithelium in two mouse strains.With this study we aim to dissect the genetic contribution to the expression of the olfactory receptor genes from environmental factors. For this, we housed a single black6 mouse with a 129S5 litter and vice versa, and dissected the MOE of the black6 animal and one 129S5 animal at 10 weeks of age. Three biological replicates were obtained for each combination. RNA was extracted using the Qiagen RNeasy kit using standard protocols. All twelve RNA samples were multiplexed together and sequenced in four lanes on the Illumina HiSeq platform.This data is part of a pre-publication release. Effects of H2be loss of function on gene expression changes in the main olfactory epithelium (MOE) as a result of activity deprivation through unilateral naris occlusion (UNO).

Project description:We have identified a replication-independent histone variant, Hist2h2be (referred to herein as H2be), which is expressed exclusively by olfactory chemosensory neurons. Levels of H2BE are heterogeneous among olfactory neurons, but stereotyped according to the identity of the co-expressed olfactory receptor (OR). Gain- and loss-of-function experiments demonstrate that changes in H2be expression affect olfactory function and OR representation in the adult olfactory epithelium. We show that H2BE expression is reduced by sensory activity and that it promotes neuronal cell death, such that inactive olfactory neurons display higher levels of the variant and shorter life spans. Post-translational modifications (PTMs) of H2BE differ from those of the canonical H2B, consistent with a role for H2BE in altering transcription. We propose a physiological function for H2be in modulating olfactory neuron population dynamics to adapt the OR repertoire to the environment. The objective of generating this dataset was to analyze the effects of H2be loss of function on gene expression changes in the main olfactory epithelium as a result of activity deprivation through unilateral naris occlusion (UNO). This dataset compares gene expression in wild type and H2be-KO main olfactory epithelium (MOE) samples from 5-week old mice that were subjected to unilateral naris occlusion (UNO) for 3 weeks starting from 2 weeks of age. Samples consist of MOE halves that were dissected and carefully removed from the medial bone. Each sample contains tissue from 2 mice (1 female and 1 male). There are three replicates for each genotype (H2be-KO or WT) and UNO side (open or closed) combination.
