Genome-wide high-throughput sequencing analysis of TLR4-overexpressing ovarian cancer cells treated with taxol and curcumin

ABSTRACT: The purpose of this study is to approach overexpression of TLR4 which contributes the drug resistance in ovarian cancer cells. However, curcumin overcomes this effects. Comparison of the transcriptional regulated genes were performed: (1) analysis of TLR4-overexpressing (T4) and mock (MCS) SKOV3 cells by taxol treatment; (2) assay of taxol alone and combined with curcumin treatments in TLR4-overexpressing (T4) SKOV3 cells. Differential expression analysis was performed with ≥ 2-fold or ≤0.5-fold (p<0.05) changed expression for pathway enrichment analysis.

A novel treatment for glioblastoma based on Taxol activation of PC1/3-depleted macrophages: evidence from a proteomic and functional study.

Project description:Tumor-associated macrophages contribute to tumor pathogenesis and represent an attractive therapeutic target. We report that the proprotein convertase PC1/3 inhibits the TLR4 Myd88-pathway induced in macrophages by the anti-cancer agent Taxol. Thus, PC1/3 knock-down in these cells exacerbates the TLR4 MyD88-dependent pathway triggered by Taxol. In PC1/3 knock-down macrophages, Taxol drives the secretion of pro-inflammatory cytokines, inhibits STAT3 signaling and counteracts tumor-supportive activities, thus inhibiting viability, growth and invasion of glioblastoma cells. Proteomic analyses indicate that their secretomes are characterized by a unique protein profile supporting a specific paracrine anti-tumoral effect. These findings unravel the potential value of a new therapeutic strategy combining PC1/3 inhibition and activation of the TLR4 MyD88-dependent pathway to switch intra-tumoral macrophages toward an anti-tumoral immunophenotype.

Expression profiling of Human SUP-T1cells under the treatment with HIV, curcumin and in combination of HIV and curcumin both.

Project description:Expression analysis was performed with human SUP-T1 cell line infected with HIV alone, treated with curcumin alone and infected with HIV in presence of curcumin. The control untreated sample was compared with all the three types of treatments to calculate the fold change. The purpose of the experiment was to identify set of genes that are dys-regulated during HIV infection and shows normal expression when treated with curcumin. This experiment is useful in identifying the mechanism of anti-HIV activity of curcumin at genetic level.

Project description:Curcumin has been shown to exhibit anti-neoplastic effects. However, due to its poor bioavailability, use of curcumin as an anti-cancer drug is limited. It is thus necessary to identify molecules that could be targeted using anti-cancer agents as alternatives to curcumin. For this, it is important to understand the underlying signaling pathways mediating the anti-neoplastic effects of curcumin. We carried out phosphoproteomic analysis (TiO2-based enrichment of Ser/Thr kinases) of head and neck cancer cell line, CAL27 treated with curcumin to identify curcumin mediated signaling pathways. This resulted in the identification of 5921 phosphopeptides (phosphosite probability≥75%) corresponding 1,878 proteins. Of these, 275 and 335 phosphopeptides corresponding to 183 and 242 proteins (≥2.0-fold) were found to be hyper- and hypo-phosphorylated respectively, in response to curcumin treatment

Phosphotyrosine profiling of curcumin-induced signaling

Project description:Curcumin, derived from the rhizome of Curcuma longa is a natural anti-cancer agent and has been shown to inhibit proliferation and survival of tumor cells. Although the anti-cancer effects of curcumin are well established, detailed understanding of the signaling pathways altered by curcumin is still unclear. In this study, we carried out SILAC-based quantitative proteomic analysis of CAL 27 cell line, a HNSCC cell line to investigate tyrosine signaling in response to curcumin. Using high resolution Orbitrap Fusion™ Tribrid™ Fourier transform mass spectrometer, we identified 627 phosphotyrosine sites mapping to 357 proteins. We observed alterations in the level of phosphorylation of 307 sites corresponding to 204 proteins upon curcumin treatment.
