Transcriptomic response of in vitro potato (Solanum tuberosum L.) to piezoelectric ultrasound

ABSTRACT: As part of a wider project to assess the impact of ultrasound on in vitro plant growth, this paper aimed to determine whether the application of piezoelectric ultrasound (PE-US) would induce changes to the transcriptome of in vitro potato (Solanum tuberosum L.). After exposing explants (single-node segments with a single leaf) to PE-US (35 kHz; 70 W) for 20 min, the effect of this stressor was determined at 0 h, 24 h, 48 h, 1 w and 4 w to assess the possible immediate and residual effects of PE-US on the potato transcriptome.

Ultrasound-activated piezoelectric nanostickers for neural stem cell therapy of traumatic brain injury therapy

Project description:Piezoelectric stimulation can promote neuronal differentiation of neural stem cells. Here we report a piezoelectric nanostickers that activate voltage-gated calcium channels (VGCC) on the surface of neural stem cells under ultrasound, inducing calcium ion influx.The influx of calcium ions leads to the upregulation of CaMKⅡ expression and phosphorylation, which initiates downstream signal transduction pathways in the process of neurogenesis. Phosphorylated CaMKⅡ (P-CaMKⅡ) enters the nucleus, where it facilities the phosphorylation of CREB. The phosphorylation of CREB enhances the expression of target gene, BDNF, ultimately leading to the completion of neurogenesis.

mRNA transcription profile of potato (Solanum tuberosum L.) exposed to ultrasound during different stages of in vitro plantlet development

Project description:Ultrasound (US) can influence plant growth and development. To better understand the genetic mechanism underlying the physiological response of potato to US, single-node segments of four-week-old in vitro plantlets were subjected to US at 35 kHz for 20 min. Following mRNA purification, 10 cDNA libraries were assessed by RNA-seq and significantly differentially expressed genes (DEGs) were categorized by gene ontology (GO) or Kyoto Encyclopedia of Genes and Genomes (KEGG) identifiers. The expression intensity of 40,430 genes from a total pool of 45,112 genes were studied. From these, several hundred genes associated with biosynthesis, carbohydrate metabolism and catabolism, cellular protein modification, and response to stress, and expressed mainly in the extracellular region, nucleus, and plasma membrane, were either up- or down-regulated in response to US. This study examines how some processes evolved over time (0 h, 24 h, 48 h, 1 week and 4 weeks) after an abiotic stress (US) was imposed on in vitro potato explants, and provides important clues to the temporal dynamics in enzyme and DEG profiles in response to this stress as the explant becomes established in vitro. Despite this abiotic stress, plantlets survived.

3D-Printed GelMA/Hydroxyapatite/Barium Titanate Piezoelectric Hydrogels for Bone Tissue Engineering

Project description:Bone defect repair remains a major clinical challenge. This study presents a novel strategy using a 3D-printed piezoelectric hydrogel scaffold—composed of gelatin methacrylate (GelMA), hydroxyapatite (HA), and barium titanate (BTO)—for functional bone tissue engineering. The GelMA/HA/BTO (GelHABT) scaffold exhibited a well-defined porous structure, enhanced mechanical stability, and, crucially, reliable piezoelectric responsiveness. This key feature enables the material to convert external mechanical stimuli, such as low-intensity pulsed ultrasound (LIPUS), into endogenous electrical signals. In vitro, the scaffold promoted BMSCs adhesion, proliferation, and osteogenic differentiation, with performance significantly enhanced under LIPUS stimulation. Mechanistic insights revealed that the piezoelectric microenvironment remodeled the cellular miRNA expression profile, particularly up-regulating osteogenesis-related miR-29b-3p and activating the AMPK signaling pathway. Collectively, this ultrasound-responsive, gene-regulating scaffold represents a promising approach for treating bone defects by leveraging piezoelectricity to actively stimulate bone regeneration.

RNA-Seq of allotetraploid wild potato Solanum acaule and autotetraploid potato Solanum tuberosum cv. Atlantic

Project description:Potato wild relatives (Solanum section Petota) are a source of genetic diversity for improving traits in modern cultivars (S. tuberosum) to meet climate challenges. Potatoes are susceptible to multiple abiotic and biotic stresses and have undergone constant improvement through breeding programs worldwide. The allotetraploid S. acaule Bitter has been used to introgress cold tolerance into potato breeding germplasm. The cold challenged transcriptome of S. acaule was compared with that of autotetraploid S. tuberosum cv. Atlantic, and was found to have fewer differentially expressed genes than the latter. Specifically, subgenome 1 has less downregulated alleles compared to subgenome 2 and S. tuberosum.

Collagen-Enhanced Piezoelectric PLLA/ZnO Microfiber Barrier Membranes for Superior Bone Regeneration

Project description:Repairing large bone defects remains a significant clinical challenge. This study explores the development of collagen-enhanced piezoelectric microfiber barrier membranes (PLLA/ZnO@COL) for bone regeneration. The membranes' physicochemical properties, especially their piezoelectric characteristics, significantly influenced cellular behavior. Scanning electron microscopy (SEM) and energy-dispersive X-ray spectroscopy (EDS) confirmed the uniform dispersion of ZnO nanoparticles within the PLLA matrix, essential for optimal piezoelectric performance. The inclusion of collagen layers enhanced the electrical properties, cytocompatibility and osteogenic activity of the membranes. In vitro analyses, including bioinformatics, revealed that PLLA/ZnO@COL membranes upregulated key osteogenic pathways such as PI3K-AKT, MAPK, and Ras, thereby enhancing osteoblast proliferation, migration, and differentiation. Low-intensity pulsed ultrasound (LIPUS) stimulation further amplified these effects, indicating a synergistic role in enhancing osteoblast functions. In vivo studies using a rat mandibular bone defect model demonstrated that PLLA/ZnO@COL membranes, with the collagen layer facing the defect, significantly fostered new bone formation compared to control groups. Micro-CT analysis at 6 and 12weeks post-implantation showed higher bone mineral density, volume, and trabecular number in the PLLA/ZnO@COL group, indicating greater osteogenic activity. In conclusion, PLLA/ZnO@COL membranes, in conjunction with LIPUS, effectively enhance osteoblast functions and induce bone regeneration, showing potential as innovative barrier membranes in bone tissue engineering. The combination of piezoelectric properties, collagen enhancement, and LIPUS stimulation presents a novel approach to improving bone healing outcomes.

Transcriptome Analyses Reveal Molecular Responses of Three Potato (Solanum tuberosum L.) Cultivars to Cold Stress

Project description:Potato (Solanum tuberosum L.), as an important food crop on the Qinghai-Tibet Plateau, is prone to low temperature and frost damage during the seedling stage, causing economic losses for farmers. In this study, transcriptome analyses were conducted on the leaves of Atlantic, KY130 and KY140 potato varieties following exposure to cold stress (CS). The genes StPAL(Soltu.Atl.09_2G005110) and StGAD(Soltu.Atl.11_3G000340), suggesting their involvement in the regulation of cold resistance in potato. “Flavonoid-related metabolism,” “lipid metabolism,” “amino acid metabolism,” “carbohydrate metabolism,” “nucleotide metabolism,” and “energy metabolism” might play an important role in the cold resistance of potato. Our results provided novel insights into the molecular mechanisms underlying cold resistance in potato.
