CircRNA-miRNA in anesthesia-induced permanent nerve injury during caesarean delivery

ABSTRACT: Permanent nerve damage caused by anesthesia is a rare but critical issue, and the mechanism of it remains unclear. Large-scale analysis of changes in noncoding RNA such as circRNA and microRNA in anesthesia-related nerve damage may provide insight into the mechanism of anesthesia-induced nerve injury.

Lumbar 5 (L5) nerve compression affects adjacent DRGs and distal nerve

Project description:Painful radiculopathy, mainly caused by physical damage to spinal nerve roots, brings algesia in their innervated areas. Intriguingly, the region of paresthesia is not always limited to the injured neuron-innervated area, and abnormal activation is also frequently observed in adjacent intact nerve segments. Since injured and uninjured nerve fibers comingle in one nerve trunk, we propose a hypothesis that the compression of the spinal nerve produces a degenerative environment at the distal end of the nerve, affecting the undamaged nerve fibers. We divided the rats into two groups: Compression of L5 spinal nerve in the experimental group and sham operation in the sham group. DRGs of L4, L5 and L6, femoral nerve and sciatic nerve were collected for RNA-seq. The results showed that L5 nerve compression resulted in inflammatory response in adjacent non-injured L4 DRG. The femoral nerve and sciatic nerve showed signs of nerve damage and repair. This suggests that injured nerve fibers can influence the properties of adjacent intact fibers through the nerve trunk which they converge.

Transcriptome analysis of the hippocampus in late postnatal mice after repeated ketamine anesthesia

Project description:Numerous recent studies showed that early anesthesia during neurodevelopment may induce changes in specific behaviors. We further studied the effects of early anesthetic exposures regarding addiction behavior later in life. Postnatal day (PND) 16 C57BL/6 mice received ketamine anesthesia for 5 consecutive days. Addiction behavior was evaluated 1 week after anesthesia treatment. Neurological changes after repeated anesthesia were evaluated using high-performance liquid chromatography, RNA sequencing, electrophysiology studies. Repeated ketamine anesthesia during the critical neurodevelopment period increased the expression of genes involved with action potential, and induced excitatory/inhibitory (E/I) imbalance in hippocampal CA1 pyramidal neurons of male mice. Such neurological changes were associated with increases in drug-induced contextual addiction memory.

CircRNA-Seq Analysis for Sciatic Nerve Injury of Rat

Project description:Here, we established a rat sciatic nerve injury model and investigated the circRNA expression profiles by Next-generation sequencing. In all, 7222 distinct circRNA candidates were found in these tissues and 4942 of these circRNAs contained at least two unique back-spliced reads. On the basis of expression of circRNA analysis, we found 131 circRNAs to be significantly (p-value<=0.05, Fold-change>=2) differently expressed in injured sciatic nerve tissues compared with matched normal tissues. All these circRNAs are derived from 21 rat chromosomes.
