ABSTRACT: Endometriotic cyst stromal cells (ECSCs) were isolated from ovarian endometriotic cyst and cultured for 48 hours in the floating three-dimensional collagen gel culture or in the conventional two-dimensional culture. Total mRNAs were extracted and subjected to gene expression microarray.

miRNA expression profiles in endometriotic cyst stromal cells (ECSCs).

Project description:To identify the miRNAs which play a role in the development of endometriosis, we explored differentially expressed miRNAs between ECSCs (N=8) and normal endometrial stromal cells (NESCs, n=8) using miRNA microarray. The expression profiles in ECSCs and NESCs were obtained, then the miRNAs were filtered by signal intensity (20.0<, at least one out of 16 samples have values within range) and flags (at least 70% of cases have good flags). The differentially expressed miRNAs in ECSCs compared with NESCs were identified using volcano plot (Fold change >= 2.0, Mann-Whiteny U test p<0.05). The primary culture of ECSCs and NESCs (controls) were utilized. Total RNA was extracted from them, and miRNA microarray analysis was performed. The miRNAs differentially expressed between ECSCs and NESCs were identified, and aberrantly expressed miRNAs in ECSCs were defined as the endometriosis related miRNAs.

The miRNA expression profiles in untreated ECSCs and decidualized ECSCs were obtained, then the miRNAs were filtered by signal intensity (50.0<, at least one out of 8 samples have values within range) and flags (at least 17% of cases have good flags). We established the criteria for regulated genes: ratio ≥2.0-fold for up-regulated genes and ratio ≤0.5 for down-regulated genes.

CCAAT/enhancer-binding protein alpha is epigenetically silenced by histone acetylation in endometriosis and promotes the pathogenesis of endometriosis: a novel therapeutic target

Project description:Accumulating evidences suggest that various epigenetic aberrations play definite roles in the pathogenesis of endometriosis. The aim of this study is to identify 1) the panel of aberrantly expressed genes that are epigenetically suppressed by histone acetylation in endometriosis; 2) the roles of CCAAT/enhancer-binding protein (C/EBP) alpha, one of the candidate molecules whose expression was epigenetically repressed in endometriotic cyst stromal cells (ECSCs), in the pathogenesis of endometriosis; and 3) the efficacy of the histone deacetylase inhibitors for the treatment of endometriosis. Subconfluent ECSCs cultured in 10-cm dish were further incubated for 72 h with or without VPA (8 mM) and/or 5aza. Total RNA from untreated ECSCs (n=4), VPA-treated ECSCs (n=4), 5aza-treated ECSCs (n=4), and VPA- and 5aza-treated ECSCs (n=4) was extracted and subjected to gene expression microarray analysis.

Transcriptome analysis of endometriotic stromal cells treated with hydrogen peroxide

Project description:In ovarian endometrioma, much iron, which is derived from blood in cyst, deposit in stroma. This iron generates oxidative stress by Fenton reaction, which is supposed to affect endometriotic stromal cells. We used gene expression microarrays to find influence of oxidative stress on endometriotic stromal cells. Gene expression microarrays revealed no statistically differences caused by oxidative stress.

Endometriotic Organoids: A Novel In Vitro Model of Endometriotic Lesion Development

Project description:The development and progression of endometriotic lesions are poorly understood, but immune cell dysfunction and inflammation are closely associated with the pathophysiology of endometriosis. A lack of suitable 3D in vitro models permitting the study of interactions between cell types and the microenvironment is a contributing factor. To address this limitation, we developed endometriotic organoids (EO) to explore the role of epithelial-stromal interactions and model peritoneal cell invasion associated with lesion development. Using a non-adherent microwell culture system, spherical organoids were generated with endometriotic epithelial cells (12Z) combined with immortalized endometriotic stromal cells (iEc-ESC) or immortalized uterine stromal cells (iHUF). Organoids self-organized with stromal cells occupying the center and epithelial cells on the periphery of the organoid. Endometriotic organoids (EO), containing iEc-ESC, resulted in the development of stratified 12Z epithelial cells compared to those with iHUF where the 12Z cells developed as a single layered epithelium. Transcriptomic analysis found 4,522 differentially expressed genes (DEG) between EO and 12Z/iHUF organoids, and the top DEG included increased expression of interleukins and prostaglandin synthase enzymes. An overlap of the EO DEG with baboon endometriotic lesions was highly significant. Finally, to mimic invasion of endometrial tissue into the peritoneum, a model was developed using EO and extracellular matrix containing human peritoneal mesothelial cells (LP9). Invasion of EO into the extracellular matrix-LP9 layer was increased in presence of estrogen or THP1-derived proinflammatory macrophages. Taken together, our results strongly support the concept that EO are an appropriate model for dissecting mechanisms that contribute to endometriotic lesion development.
