RNA Sequencing of C elegans control and mutant worms after 24 hour P.aeruginosa infection

ABSTRACT: Purpose: After infection there was significant decrease in the lifespan of mutant animals . To decipher the mechanism responsible and the changes in the gene expression of mutant worms Methods: Control and mutant worms were exposed at young adult stage to P.aeruginosa PA14 infection for 24 hours. Total RNA was extracted and RNA sequencing was done following standard protocols Results: Detailed analysis of the transcriptomic data show that immune genes were significantly upregulated which have role in lifespan Conclusions: This study showed the role of pathogenic infection in the life span modulation

Quantitative Analysis of Worms after P. aeruginosa Infection

Project description:To clarify the mechanisms of innate immune responses mediated by epithelial barrier disruption. Reveal the host’s broad transcriptional response to P. aeruginosa infection during disruption of the intestinal barrier. We used RNA-Seq analysis to study the transcriptomic profiles of wt worms were exposed to either P. aeruginosa PA14 or E. coli OP50 for 24 hours, and the transcriptomic profiles of egl-44(MT2247) mutants fed E. coli OP50 for 24 hours. Differential expression analysis was performed by comparing wt worms exposed to PA14 versus wt worms exposed to E. coli OP50, and egl-44(MT2247) mutants fed E. coli OP50 versus WT worms fed E. coli OP50.

Gene expression in the small intestine of mice colonized with Pseudomonas aeruginosa

Project description:Celiac disease (CeD) is a food sensitivity characterized by a breakdown of oral tolerance to gluten proteins in genetically predisposed individuals, although the underlying mechanisms are incompletely understood. To evaluate a functional role of bacterial proteases in CeD, we colonized germ-free or clean SPF mice with P.aeruginosa PA14 or P.aeruginosa PA14 disrupted in lasB gene (elastase). We show gluten-independent, PAR-2 mediated upregulation of inflammatory pathways by LasB in C57BL/6 mice without villus blunting. In mice expressing CeD risk genes, P. aeruginosa elastase synergized with gluten to induce more severe inflammation that was associated with moderate villus blunting.

Transcriptional responses of K. pneumoniae, S. maltophilia, P.aeruginosa, and B. thailandensis exposed to lung surfactant

Project description:The interactions between Gram-negative respiratory pathogens and the host environment at the site of infection largely unknown. Pulmonary surfactant serves as an initial point of contact for inhaled bacteria entering the lung and is thought to contain molecular cues that aid colonization and pathogenesis. To gain insight into this ecological transition, we characterized the transcriptional responses of Pseudomonas aeruginosa PA14, Burkholderia thailandensis E264, Klebsiella pneumoniae MGH 78578, and Stenotrophomonas maltophilia K279A exposed to purified pulmonary surfactant (Survanta) through microarrays. This study provides novel insight into the interactions occurring between Gram-negative opportunistic pathogens and the host at an important infection site, and demonstrates the utility of purified lung surfactant preparations for dissecting host-lung pathogen interactions in vitro. The goal of this study was to compare the transcriptional responses of Pseudomonas aeruginosa PA14, Burkholderia thailandensis E264, Klebsiella pneumoniae MGH 78578, and Stenotrophomonas maltophilia K279A exposed to pulmonary surfactant using a custom affymetrix chip designed for their genomes. ChIPseq examination of EAP-1 binding across generations of spr-5(by101) mutant worms

Project description:How epigenetic information is transmitted from generation to generation remains largely unknown. Deletion of the C. elegans Histone H3 lysine 4 dimethyl (H3K4me2) demethylase spr-5 leads to inherited accumulation of the euchromatic H3K4me2 mark and progressive decline in fertility. Here we identified a genetic network of chromatin-modifying factors, including the H3K4me1/me2 methyltransferases SET-17 and SET-30, the H3K9me1/me2 methyltransferase MET-2, an H3K9me3 methyltransferase, SET-26, the H3K9me3 demethylase JMJD-2, and an H3K9me reader EAP-1, which regulate the trans-generational flow of epigenetic information. Importantly, genetic ablation of set-17, set-30, jmjd-2, or eap-1 suppresses the progressive transgenerational phenotypes, while loss of SET-26 or MET-2 accelerates the infertility of spr-5 mutant worms. We further show that loss of spr-5 also causes a trans-generational increase in lifespan, which is dependent on these chromatin regulators as well as DAF-36 and DAF-12, which control a germline to soma longevity signaling pathway. These findings suggest that the balance between the euchromatic H3K4 and the heterochromatic H3K9 methylation regulates trans-generational effects on longevity and fertility. EAP-1 binding ChIPseq libraries were prepared from 50 ul of packed young adult worms which were maintained at 16 degrees until the appropriate generation and then shifted to 25 degrees after birth. Two biological repeats were generated for wildtype and generation 20 spr-5(by101) mutant worms and a single repeat for generation 10. There were four input samples and eap-1 null mutant worms were also analyzed.
