Project description:To evaluate the change of gene expression at cerebral infarction by the treatment of IV-human umblical cord derived mesenchymal stem cell. RNA was isolated from the ipsilateral hemisphere to MCAo in rats. At 72 h post-MCAo, the ipsilateral hemisphere subjected to MCAo was used for mRNA microarray. RNA was isolated from the ipsilateral hemisphere to MCAo in rats without MCAo (control group, n=5), rats treated with 1x106 IV-hUMSC (hUMSC group, n=6) and saline (saline group, n=5) at 24h post-MCAo.

Project description:Middle cerebral artery occlusion (MCAo) in rat represent the ischemic stroke in human. Rodents subjected to MCAo and treated with venom phospholipase A2 showed reduction in infarct volume after 24hours of stroke. We studied the global gene expression of the reduction in infarct volume using Affymetrix Gene Chips. We analysed all the genes that were up or down regulated in the study. Total RNA isolated from sham, MCAo and MCAo+nPLA rat brains, was pooled to minimize inter-individual variation and hybridized to each array of the RAE-230A or U34A GeneChipTM according to protocols described in the GeneChipTM expression analysis package (Affymetrix, CA).

Macaca Fascicularis Differential Gene Expression Following Stroke: Control versus Tat-NR2B9c Treated Cortex at 1 hour and 6 hours following middle cerebral aretry occlusion (MCAO)

Project description:Differential gene expression comparison between cortex of placebo and PSD-95 inhibitor (Tat-NR2B9c), treated cynomolgus macaques at 1 and 6 hours after middle cerebral artery occlusion. The purpose of this study was to identify changes in differential gene expression in the cortex of animals receiving strokes across the two time points and bewtwee the two treatment groups. Differential gene expression was defined as the ratio of fluroescene detected for expressed sequence tags in the left, unoperated cortex versus the contralateral, right, ischemic cortex at each time point. Two colour, Agilent, 4x44k whole genome macaca mulata microarrays with sample rna hybridized to Cy5 channel and universal macaca mulatta rna control on the Cy3 channel were obtained for each hemisphere. Comparisons between treatment groups at each time point and within in each treatment group across each time point were made to detect differential gene expression. Significant differential gene expression was defined as greater than 2-fold change and p<0.05 by T-test. Three condition experiment comparing the effect of treatment with placebo versus Tat-NR2B9c administered 5 min after MCAO with tissue samples collected 1h and 6h after MCAO and a second comparison of gene expression within each treatment group between 1h and 6h: 3 biological replicates in each group

Project description:Group 2 innate lymphoid cells (ILC2s) are key players in immune regulation and tissue repair in barrier immunity, but they are rarely seen in the brain. Since ILC2s can be intrinsically expanded by IL2/IL2 antibody complex, we investigated the transcriptomic changes induced by IL2/IL2 antibody complex (IL2-JES6-1) after ischemic stroke, we performed single-cell RNA sequencing of the CD45high immune cells from the infarcted hemisphere at day 14 after MCAO using 10x Genomics scRNAseq. To investigate the key mechanism of ILC2s-mediated long-term recovery after ischemic stroke, we performed single-cell RNA sequencing of the infarcted hemisphere at day 14 after MCAO using 10x Genomics scRNAseq.
