PRD1-BF1 drives VEGF-mediated innate immune suppression in tumor vasculature

Project description:Our experiments have demonstrated that VEGF signalling is a critical component of oncolytic virus infection in tumor vasculature. This effect is in part mitigated by downstream activation of the master immunoloigcal transcriptional repressor known as PRD/BF1. To investigate the role of the VEGF-PRD/BF1 signalling axis in the context of oncolytic virus infection, we isolated the RNA from sub-confluent human umbilical vein endothelial cells treated under four conditions (siRNA Control, siRNA Control + WyTK-, siRNA Control + WyTK- + VEGF165 and siRNA PRD/BF1 + WyTK- + VEGF165. Using the RNA pooled from four experiments, the goal of this study was to determine the nature & fraction of genes repressed by VEGF in the context of infection, which could be rescued by PRD/BF1.

Microenvironmental-derived Regulation of HIF-Signaling Drives Transcriptional Heterogeneity in Glioblastoma Multiforme

Project description:The evolving and highly heterogeneous nature of malignant brain tumors underlies their limited response to therapy and poor prognosis. In addition to genetic alterations, highly dynamic processes such as transcriptional and metabolic reprograming play an important role in the development of tumor heterogeneity. The present study reports an adaptive mechanism in which the metabolic environment of malignant glioma drives transcriptional reprogramming. Multi-regional analysis of a glioblastoma patient biopsy revealed a metabolic landscape marked by varying stages of hypoxia and creatine enrichment. Creatine treatment and metabolism was further shown to promote a synergistic effect through up-regulation of the glycine-cleavage system and chemical regulation of Prolyl-Hydroxylase Domain (PHD). Consequently, creatine maintained a reduction of reactive oxygen species and change of the a-ketoglutarate/succinate ratio leading to an inhibition of the HIF-signaling in primary tumor cell lines. These effects shifted the transcriptional pattern toward a proneural subtype and reduced the rate of cell migration and invasion in vitro.

GDNF/GFRA1 signaling contributes to chemo- and radioresistance in glioblastoma

Project description:Glioblastoma (GBM) is the most common primary brain tumor in adults, characterized by an inherent aggressivity and resistance to treatment leading to poor prognoses. While some resistance mechanisms have been elucidated, a deeper understanding of these mechanisms is needed to increase therapeutic efficacy. In this study we first discovered glial-cell derived neurotrophic factor (GDNF) to be upregulated in patient-derived glioblastoma spheroid cultures after chemotherapeutic temozolomide (TMZ) treatment, through RNA-Seq experiments. Therefore, we investigated the role of the GDNF/GDNF receptor alpha 1 (GFRA1) signaling pathway as a resistance mechanism to chemotherapy with temozolomide and lomustine as well as irradiation using patient-derived glioblastoma spheroid cultures. With qPCR experiments we showed a consistent upregulation of GDNF and its primary receptor GFRA1 following all three lines of treatment. Moreover, CRISPR/Cas9 knock-outs of GDNF in two patient-derived models sensitized these cells to chemotherapy treatment, but not radiotherapy. The increased sensitivity was completely reversed by the addition of exogeneous GDNF, confirming the key role of this factor in chemoresistance. Finally, a CRISPR KO of GFRA1 demonstrated a similar increased sensitivity to temozolomide and lomustine treatment, as well as radiotherapy. Together, our findings support the role of the GDNF/GFRA1 signaling pathway in glioblastoma chemo and radioresistance.
