Project description:We use RNA-sequencing to measure global transcription in uteri of young and aged (~1yr old) C57BL/6 females at day 3.5 (E3.5) of pregnancy, and of the decidual part of E11.5 placentas developed in young and aged (~1yr old) females. We report dys-regulation of gene expression in the E3.5 uteri and E11.5 deciduae of aged mice. We compare the transcriptome of young and aged E11.5 deciduae with that of young deciduae from E9.5-E12.5 of pregnancy, and report that aged E11.5 deciduae are developmentally retarded in comparison to young deciduae.

Competing endogenous RNA expression profiling in decidua after LPS treatment

Project description:To investigate the competing endogenous RNA expression changes in decidua under the inflammatory reaction, LPS (100 μg/kg body weight) was intraperitoneally injected into the mice at 15 days of pregnancy. Premature births has been found after approximately 48 h of treatment. When bleeding found in vagina, the decidua were selected in asepsis condition, and total RNA was isolated to analyze the expression of competing endogenous RNA compared with cesarean sections.

Global transcriptome analysis of mouse uterus during embryo implantation by SAGE

Project description:The aim of this study was to examine differential gene expression profile in mouse uterus on day 5 of pregnancy between implantation sites and inter-implantation sites. There are 104,520 tags sequenced and acquired, 51,306 for non-implantation site, and 53,214 for implantation site. Keywords: Implantation, Uterus, Differential gene expression, Implantation sites and inter-implantation sites were isolated from the mouse uteri on day 5 of pregnancy from at least 20 mice. Implantation sites were identified by tail-vein injection of 0.1 ml of 1% Chicago blue 5 min before sample collection. SAGE analysis was used to examine differential gene expression in mouse uterus during embryo implantation.

This study has built a meaningful basis for future investigation in elucidating the molecular nature of maternal-fetal interactions during pregnancy establishment and maintenance. Project description:Implant failure and insufficient placental development are important causes of female infertility, recurrent miscarriage, and other pregnancy-related problems. A better understanding of gene expression profiling based on high-throughput sequencing technology is important for the study on the development of the placenta and the causes of pregnancy-related diseases. In this study, we collected 6 first-trimester chorionic villus and decidua tissues and obtained the transcriptome database on high-throughput sequencing. We performed routine principal component analysis (PCA) on these 12 samples and identified differentially expressed genes from samples with different sex background. And identified genes highly expressed in villus and decidua, respectively.
