Profiling circulating microRNA expression in experimental sepsis by cecal ligation and puncture

Project description:To profile the expression of circulating microRNAs (miRNAs) of mice in experimental sepsis by cecal ligation and puncture (CLP), the whole blood samples were obtained from C57BL/6 mice at 4, 8, and 24 h following CLP for miRNA expression analysis using a miRNA array (The Mouse & Rat miRNA OneArray® v3). Briefly, mice were anesthetized with a combination of ketamine and xylazine as the anesthetic/analgesic agents and a midline abdominal incision was made. The cecum was mobilized, ligated in the middle of cecum below the ileocecal valve, punctured once with a 21 G needle, and a little stool was squeeze out of the cecum to induce polymicrobial peritonitis. The abdominal wall was closed in two layers. Sham-operated mice underwent the same procedure, including opening the peritoneum and exposing the bowel, but without ligation and needle perforation of the cecum.

Effect of LXR agonist T0901317 on the intestinal transcriptome in mice.

Project description:Mice were treated for three days by oral gavage with the synthetic LXR ligand T0901317 (Cayman chemical, USA) using a dose of 50 mg/kg/day in vehicle (0.5% Carboxy-methylcellulose; 400 cPs (Sigma-Aldrich, USA); 0.2% Tween80 (Sigma-Aldrich, USA)) or solely the vehicle (control group). Mice were anesthetized with isoflurane and euthanized by cardiac puncture followed by cervical dislocation. Intestines were flushed with ice cold PBS and frozen in liquid nitrogen.

Reversibility of etomidate-induced contractile arrest through activation of myocardial autophagy

Project description:Anesthetic management of heart failure patients undergoing noncardiac surgery remains challenging due to cardiac suppressive properties of anesthetics. Due to varying electrophysiological properties, small and large animals are not good models for studying human myocardial anesthetic responses. Here we use hypertrophic (HCM), dilated (DCM) and healthy human induced pluripotent stem cells derived cardiomyocytes (hiPSC-CMs) to evaluate the cardiac suppression of propofol and etomidate. We demonstrate that propofol and etomidate act through GABAA receptors and contractile inhibition can occur without cell-cell junction. At supraphysiological dosage (100mM), we discovered that cardiac suppression induced by etomidate is reversible while propofol is not. Using transcriptome profiling, we uncover that etomidate was capable of inducing autophagy, likely through induction of cytosolic calcium. Lack of autophagy induction in propofol treated cardiomyocytes were associated with increased apoptosis. Together, we provide the robustness of using hiPSC-CMs as an in vitro cardiotoxicity platform for anesthetics. To delineate how etomidate confers cardioprotection during contraction inhibition, we performed transcriptome profiling on DCM hiPSC-CMs treated with either 10 μM propofol, 10 μM etomidate or DMSO control.

Project description:Mice underwent cecal ligation and puncture (19 GA with concurrent administration of imipenem), sham operation, or were unoperated. 14 days after operation, microglia were isolated by fluorescent activated cell sorting for CD11b+/CD45mid/CD64+ cells. For each sample, the brains of 3 mice were pooled. RNA was then isolated for transcriptome analysis.
