Project description:Willow (Salix), a dioecious plant, is an important ornamental tree species in the world. Salix linearistipularis, a perennial woody plant species naturally distributed on the Songnen Plain saline-alkali land in northeast China, has a high saline condition. To study the sexual differences of S. linearistipularis in salinity tolerance, the physiological and transcriptional responses to salinity were compared between female and male cuttings. Under salinity stress, the female leaves exhibited higher superoxide dismutase and peroxidase activities and photosynthetic capacity, and lower H2O2 contents than those of male leaves. Under salinity stress, sodium (Na+) accumulation in female leaves was lower than that in the male leaves. The non-invasive micro-test showed that the net Na+ efflux in the salt-treated female roots was higher than that in male roots. Physiological responses revealed that female cuttings were more tolerant than males, which may be mainly due to females having lower leaf Na+ accumulation and higher root Na+ efflux capacity than males. Transcriptional analyses showed that 108 differentially expressed salt-responsive genes were identified in both female and male roots; most of these showed sexual differences in expression patterns under salinity stress. RNA-seq combined with qPCR analysis showed that the salt-induced expression of four Na+/H+ antiporter (NHX) genes (SlNHX3, 5, 6, 7) in female roots was higher than that in male roots. Transcriptional analyses revealed that the higher Na+ efflux capacity in female roots than in male roots may be closely related to the differential expression of salt-responsive genes, especially NHX genes.

Those up-regulated during the NaCl treatment may protect the plants from damages caused by salt stress. 2 month-old S. matsudana plants which were treated with 100 mM NaCl and control plants were used for RNA extraction and hybridization on Affymetrix microarrays. We sought to obtain salt responsive genes that protect the plants from stress injury.Those differentially expressed genes identified by the microarray would help to understand the mechanism of S. matsudana reacting to salt stress.

Project description:Hernandia nymphaeifolia is an endangered mangrove associate with high ecological, ornamental, and medicinal values. Its special combination of glycophytic and halophytic characteristics provides an ideal system for investigating salt-responsive mechanisms that may be easily extended to genetic improvement of crops with high salt tolerance. However, little is known regarding salt-responsive mechanisms in H. nymphaeifolia. In this study, we posed gradient salt treatments on H. nymphaeifolia seedlings and investigated their physiological and transcriptional reprogramming in response to salinity stress. The results revealed that hyper-salinity stress posed more adverse impacts on leaf growth, cell integrity and photosynthetic performance of H. nymphaeifolia seedlings compared to those in growing in fresh water or low salt condition, mirroring its nature as a mangrove associate. Genes associated with osmolarity sensor and regulator, ROS scavenging and ion homeostasis were differentially expressed accordingly to alleviate the destructive effects. Furthermore, our results unraveled some pivotal kinases and proteins that regulate the synergistic expression of salt-responsive genes, which may act a key role in regulating the transcriptional remodeling upon salinity stress. These findings enrich our knowledge on the molecular mechanisms underlying the phenotypic plasticity of mangrove associates, and also provide valuable genetic resources for mangrove’s conservation and potential bioengineering application in agricultural field.

Project description:Few phytoremediation studies have been conducted under semi-arid conditions where plants are subjected to drought and/or salinity stress. Although the genus Salix is frequently used in phytoremediation, information regarding its tolerance of drought and salinity is limited. In the present study, Salix acmophylla Boiss. cuttings from three sites (Adom, Darom and Mea She'arim) were tested for tolerance to salinity stress by growing them hydroponically under either control or increasing NaCl concentrations corresponding to electrical conductivities of 3 and 6 dS m-1 in a 42-day greenhouse trial. Gas exchange parameters, chlorophyll fluorescence and concentration, and water-use efficiency were measured weekly and biomass was collected at the end of the trial. Root, leaf and stem productivity was significantly reduced in the Adom ecotype, suggesting that Darom and Mea She'arim are the more salt-tolerant of the three ecotypes. Net assimilation and stomatal conductance rates in salt-treated Adom were significantly reduced by the last week of the trial, coinciding with reduced intrinsic water use efficiency and chlorophyll a content and greater stomatal aperture. In contrast, early reductions in stomatal conductance and stomatal aperture in Darom and Mea She'arim stabilized, together with pigment concentrations, especially carotenoids. These results suggest that Darom and Mea She'arim are more tolerant to salt than Adom, and provide further phenotypic support to the recently published data demonstrating their genetic similarities and their usefulness in phytoremediation under saline conditions.
