Project description:Plant hormones and small secretory peptides often function as environmental stress mediators. Some recent reports indicate that small secretory peptides, such as CLAVATA3/EMBRYO SURROUNDING REGION-RELATED (CLE), also function as mediators of environmental stimuli. CLE３ is induced in roots by defense elicitor treatment. Plants without functional CLE3 showed compromized defense gene responses in shoots when plant roots were treated with NaSA. Here, we identified specific genes downstream of CLE3 in roots and shoots with transformed Arabidopsis plants.

Project description:Our previous study on CBP60g, a calmodulin binding protein that is important for disease resistance and microbe-associated molecular pattern (MAMP)-induced SA accumulation, led to our discovery of a closely related family member CBP60h. CBP60h is also important for defense against P. syringae but is induced differently by pathogen and MAMP stimulus. Transcriptome profiling of cbp60h mutants suggested that CBP60h might be primarily functioning in response against P. syringae. We constructed a double mutant of cbp60g and cbp60h, which demonstrated severely defective defense against P. syringae and SA accumulation. Profiling of the cbp60g/h showed that its expression pattern is very similar to that of pad4. Transient expression in Tobacco showed that both CBP60g and CBP60h localized to nucleus. Our observation suggest that CBP60g and CBP60h share partially redundant but critical role in defense response and SA signaling. This experiment consists of three biological replicates. For each genotype, two leaves per plant were pooled from three pots to prepare total RNA.

Project description:Arabidopsis nuclear RecA homologue RA51D were reported to be involved in plant defense responses. Plant organelles such as mitochondria and chloroplast also have their own RecA homologues. We focused on chloroplast RecA homologue RECA1 because it has been well known that the precursors of phytohormones and secondary metabolites related to plant defense responses are synthesized in chloroplast and recent studies have identified several chloroplastic proteins invoved with plant defense responses. We used microarrays to investigate the global gene expression changes by RECA1 and identified the up-regualted genes involved with plant defense response. 2-week-old Col-0 and RECA1(at1g79050)-overexpressing plants without any treatments were used for RNA extraction and hybridization on Affymetrix microarrays. Col-0 plants were used as control and T3 RECA1-overexpressing transgenic plants were used in this study.

Project description:Accumulating information indicates that plant disease resistance signaling pathway frequently interact with other pathways regulating developmental processes or abiotic stress responses. However, the molecular mechanisms of these types of crosstalk remain poorly understood in most cases. Here we report that OsWRKY13, an activator of rice resistance to both bacterial and fungal pathogens, functions as a convergent point of the crosstalk between pathogen-induced salicylate-dependent defense pathway and at least five other physiological pathways. Genome-wide analysis of the expression profiling of OsWRKY13-overexpressing lines showed that OsWRKY13 directly or indirectly regulates the expression of more than 500 genes, which are potentially involved in different physiological processes according to the classification of Gene Ontology database. Comparing the expression patterns of genes functioning in known pathways or cellular processes upon pathogen infection and the phenotypes between OsWRKY13-overexpressing and susceptible wild-type plants, we find that OsWRKY13 is also regulator of other physiological processes during pathogen infection. OsWRKY13-involved disease resistance pathway synergistically interacts with glutathione/glutaredoxin system and flavonoid biosynthesis pathway via OsWRKY13 to monitor redox homeostasis and may enhance the biosynthesis of antimicrobial flavonoid phytoalexins. Meanwhile, OsWRKY13-invloved disease resistance pathway antagonistically interacts with SNAC1-mediated abiotic-stress defense pathway, JA signaling pathway, and terpenoid metabolism pathway via OsWRKY13 to suppress salt and cold defense responses as well as may retard rice growth and development. Experiment Overall Design: To determine the genes directly or indirectly regulated by OsWRKY13 genome-widely, we performed the microarray analysis using the Affymetrix Genechip Rice Genome Array containing total 57381 probe sets. Two independent OsWRKY13-overexpressing rice lines, D11UM 1-1 and D11UM 7-2 (Qiu et al., 2007), and wild-type Mudanjiang 8, which was susceptible to Xoo and M. grisea, were used for the analyses.The experiment was repeated three times.

Project description:The goal of the RNA seq was to investigate the transcriptome changes induced by Pseudmonas syringae pv. tomato J4 in wild-type tomato 'Moneymaker' and transgenic 'Moneymaker' overexpressing the Arabidopsis ELP4 (AtELP4) gene. Results showed that P. syringae pv. tomato J4 induced dramatic transcriptional changes in both the wild-type and transgenic tomato plants. Interestingly, a group of defense genes including PR-5x, Pti5, PR1b1, and CHI3/9/14/17, which are associated with resistance to the hemibiotrophic bacterial pathogen Ralstonia solanacearum, were induced to higher levels in the AtELP4 transgenic tomato than in the wild type at 8 and 24 hr after P. syringae pv. tomato J4 infection. These results indicate that overexpression of AtELP4 in tomato leads to faster and/or stronger induction of some defense genes.

Project description:Plant immune responses to pathogen attack involve various defense mechanisms and among them, the Hypersensitive Response (HR), a form of programmed cell death occurring at invasion sites. AtMYB30, a transcription factor acts as a positive regulator of a cell death pathway conditioning the HR. We show by microarray analyses of Arabidopsis plants misexpressing AtMYB30 that the genes encoding the four enzymes forming the acyl-coA elongase complex, responsible for very long chain fatty acids (VLCFA) biosynthesis, are putative targets.
