Schwann Cell Tn5Prime

ABSTRACT: Here we provide sequencing data derived from our efforts to identify SOX10-regulated promoters in Schwann cells genome-wide. We assess the activity of SOX10-bound promoters using Tn5Prime to identify and quantify transcription start sites in adult peripheral nerve, in differentiating primary Schwann cells, and upon ablation of SOX10 in vitro. Tn5Prime libraries were prepared essentially as described by Cole and colleagues (NAR, 2018). Analyses focused on transcription start sites associated with H3K4me3 and SOX10 ChIP-Seq peaks from sciatic nerve (ChIP-Seq data derived from previously published datasets).

Genome-wide identification of target genes for the transcription factor Sox10 in the Msc80 Schwann cell line.

Project description:To identify target genes of the Sox10 transcription factor in peripheral glia in a genome-wide and unbiased approach, we performed CRISPR/Cas9-dependent genome editing of the mouse Sox10 gene in the Msc80 Schwann cell line, followed by RNA-Seq studies on Sox10-expressing control Msc80 cells and Sox10-negative genome edited cell clones. Among others we detected substantial changes in the expression of genes associated with Schwann cell adhesion and the formation of the node of Ranvier.

RNA-seq analysis of S16 Schwann cells treated with siRNA for Nr2f1/Nr2f2 (CoupTFI/CoupTFII)

Project description:In a previous study, we had compared SOX10-bound regulatory elements in Schwann cells and oligodendrocytes, and we identified a specific enrichment for nuclear receptor motifs at SOX10 binding sites in Schwann cells. We initially focused on NR2F1 and NR2F2 (CoupTF1/CoupTF2) since they are expressed from neural crest through Schwann cell maturity, and found that knockdown of nuclear receptors Nr2f1 and Nr2f2 in primary Schwann cells downregulated genes such as Myelin Basic Protein (Mbp) and Desert Hedgehog (Dhh). In this study, we have elucidated a NR2F-regulated target gene network in Schwann cells, which revealed enrichment for non-myelinating Schwann cell genes. Cut&Run assays in S16 Schwann cells revealed novel, genome-wide binding sites of NR2F1/2 and downstream transcription factors, Retinoid X Receptor (RXRG) and TEA-Domain factor (TEAD1).

Histone H3K4 trimethylation in rat peripheral nerve

Project description:ChIP-seq of H3K4me3 in rat peripheral nerve was used to identify transcription start sites associated with Schwann cell-expressed genes. The analysis was performed in injured and control nerve to identify injury-responsive changes in Schwann cells. H3K4me3 ChIP samples were prepared from rat sciatic nerve at 1 day post-transection using both the distal stump of the injured nerve and the contralateral (sham) nerve.

Project description:Peripheral glial Schwann cells switch to a repair state after nerve injury, proliferate to supply lost cell population, migrate to form regeneration tracks, and generates a permissive microenvironment for nerve regeneration. Exploring essential regulators of the repair responses of Schwann cells may benefit the clinical treatment for peripheral nerve injury. In the present study, FOSL1 regulates Schwann cell phenotype modulation and provided a novel therapeutic approach to orchestrate the regeneration and functional recovery of injured peripheral nerves.

Altered genomic occupancy of H2A.Z in early postnatal mouse sciatic nerve after Schwann cell-specific deletion of the Ep400 chromatin remodeler

Project description:To address the role of INO80/SWR-type remodeling complexes, we deleted Ep400 in developing Schwann cells. Ep400 is involved in exchanging H2A-containing standard with H2A.Z-containing nucleosomes. Ep400 becomes important for proper Schwann cell development at differentiation. Its absence in Schwann cells leads to a peripheral neuropathy. ChIP-Seq studies were performed with H2A.Z-specific antibodies on sciatic nerve from control mice and mice with a Schwann cell-specific deletion of Ep400 to determine the genomic occupancy of this histone variant. One group of genes with strongly altered H2A.Z occupancy around the transcriptional start site consisted of developmental regulatory genes whose occurrence and action is normally limited to earlier stages of development.
