Effects of newly developed pulp-capping materials on early response of stem cells from exfoliated deciduous teeth: cell differentiation, migration, and proliferation based on next gene sequencing analysis


ABSTRACT: Aim : This study aimed to identify early effect of newly developed pulp capping materials on differentiation, migration, and proliferation of stem cells from exfoliated deciduous teeth (SHED) using next gen sequencing analysis. Methodology : SHED were cultured and exposed to MTA, Biodentine (BD), and Theracal (TC) for 24 and 72 h. WST assay was performed to evaluate cell viability. After total RNA was isolated, QuantSeq 3’ mRNA-Sequencing was performed to identify differential gene expression in MTA-, BD-, TC-exposed SHED. Bioinformatics analysis was also performed. Results : WST assay showed no significant difference in SHED viability among materials (p>0.05). In gene set enrichment analysis, hallmark gene set enrichment differed. Gene sets associated with epithelial mesenchymal transition and hedgehog signaling were enriched in all materials. In BD, gene sets with inflammatory response and Wnt/β-catenin signaling were enriched, whereas enrichment of gene sets with TNF-α signaling and apoptosis were identified in TC. Conclusions : Despite cell viability of SHED exhibited similar results among MTA, BD, and TC, patterns of gene networks differed, suggesting that diverse gene functional difference may be involved in these materials.
