Project description:Hepatic stellate cells(HSCs) are the main effector cells of liver fibrosis. In order to study the effect of mesenchymal stem cells(MSCs) on microRNAs expression of HSCs, we co-cultured HSCs LX-2 activated by TGFβ1 with human umbilical cord MSCs(hUC-MSCs) for 48 hours, and compared the differentially expressed miRNA with LX-2 cultured alone by high-throughput sequencing. The results showed that two mature microRNAs expressed increased, and nine expressed decreased.

Project description:To investigate whether Aramchol might reduce liver fibrosis in part directly inhibiting HSCs, we treated LX-2 cells with Aramchol, 10 µM for 24 or 48 hours, then performed RNA sequencing. Sequencing libraries were made with the TruSeq Stranded mRNA Library Prep Kit by manufacturers suggestions. Sequencing parameters are as follows: poly-A RNA enrichment, 175 bp paired reads ran on HiSeq2500 Illumina instrument. Mapping of raw sequencing reads was performed using STAR (2.4.0c) to the UCSC GRCh37/hg19 reference genome.

Furthermore, we analyzed the transcriptome of oxo-C14-HSL pretreated plants after a secondary challenge with 100 nM flg22 for 2 and 24 hours after treatment.

The transfection with the expresion plasmid was unsucessful so this empty vector data has been used alone to simply examine the effect of TGFβ1 treatment on A2058 cells.
