Project description:Explore in vivo ovarian follicle transcriptomics from primordial follicles to the antral stage. Ovarian follicles were isolated from CD-1 mice. Entire ovaries were collected at post-natal day 3 and day 4 to collect primordial follicles. Primary follicles (70-90 µm in diameter), two-layered secondary follicles (100-130 µm), multi-layer secondary follicles (150‒180 µm), and pre-antral follicles (200‒300 µm) were mechanically isolated from post-natal day 10, 12, 16, and 18 ovaries, respectively. Antral follicles (400‒600 µm) were mechanically isolated from pregnant mare serum gonadotropin (PMSG)-treated mice ovaries at post-natal day 20. Follicles were then aspirated and combined per ovarian follicle maturation stage (e.g., primary, two-layered secondary). Three different samples were collected from each pooled follicular stage for transcriptomic analysis. RNA was purified and hybridized in MouseRef-8 v2.0 Expression BeadChip Kit (Illumina, San Diego, CA), as previously described (Skory, Bernabe et al., 2013).

Project description:Women are born with thousands of follicles in their ovaries but less than 1% will produce a mature oocyte. What distinguishes the 99% of ovarian follicles destined for demise from the rare 1% that succeed? Here, we leveraged a unique clinical opportunity, paired follicular fluid samples from small antral follicles containing viable and atretic oocytes, to uncover the proteomic determinants governing oocyte fate. Our analysis identified nearly 1,500 proteins, revealing that healthy follicular fluid is enriched with proteins crucial for energy metabolism, antioxidative defense, structural integrity, and robust signaling networks, supporting successful oocyte survival and maturation. In contrast, atretic follicles exhibited a distinct inflammatory signature characterized by acute-phase and immune activation proteins, indicative of active degeneration. These novel proteomic insights deepen our understanding of follicle biology and open new avenues for biomarker development, optimization of assisted reproductive technologies, and targeted therapeutic strategies to preserve and enhance female fertility.

Using Hi-C (genome-wide chromatin conformation capture) and Smart RNA-seq of oocytes from 6- weeks or 10- months aged ovaries, the abnormal loose chromatin structures and disturbing expression of meiosis associated genes at metaphase I phase were disclosed. Furthermore, the transcriptomic landscape of granulosa cells (GCs) surrounding oocytes from young and aged ovaries reveled that oocyte meiotic maturation was accompanied with a robust increased expression of genes involved with the mevalonate (MVA) pathway in GCs from young ovaries but these genes expression was not upregulated to counterpart level in GCs from aged ovaries.The inhibitor of MVA pathway of GCs, Statins significantly decreased polar body extrusion rate and increased the rate of irregularly assembled spindles and misaligned chromosomes remarkably in oocytes of culumus-oocyte complex (COCs) from young ovaries . Correspondingly, the activitor of MVA pathway of GCs, Geranylgeraniol ameliorated ovarian reserve and reduced meiotic defects in oocytes of COCs from aged ovaries. Mechanistically, MVA pathway activation in GCs culminated oocyte meiotic maturation by upregulating EGF signaling via LH receptor on GCs surrounding oocytes. Together, MVA pathway is a promising therapeutic target for prompting quality of oocytes from aged ovaries.

Project description:The mechanisms contributing to age-related deterioration of the female reproductive system are complex but aberrant protein homeostasis is a major contributor. We elucidated the exceptionally stable proteins, structures, and macromolecules that persist in ovaries and gametes throughout the reproductive aging timeline in mammals. Ovaries exhibit localized structural and cell-type specific enrichment of stable macromolecules throughout both the follicular and extrafollicular environments. Moreover, both ovaries and oocytes harbor a panel of exceptionally long-lived proteins, including cytoskeletal components, mitochondrial, and oocyte-derived proteins. The exceptional persistence of these long-lived molecules might play a critical role in both lifelong maintenance and age-dependent deterioration of reproductive tissues.

Project description:EXOSC10 is a catalytic subunit of the nuclear RNA exosome with an exoribonuclease activity. The enzyme processes and degrades different classes of RNAs. To delineate the role of EXOSC10 during oocyte growth, specific Exosc10 inactivation was performed in the oocytes from the primordial follicle stage onward using the Gdf9-iCre; Exosc10f/- mouse model (Exosc10cKO(Gdf9)). Exosc10cKO(Gdf9) female mice are infertile. The onset of puberty and the estrus cycle in mutants are initially normal and ovaries contain all follicle classes. By the age of eight weeks, vaginal smears reveal irregular estrus cycles and mutant ovaries display a complete depletion of follicles. Mutant oocytes retrieved from the oviduct are degenerated, sometimes showing an enlarged polar body which may reflect a defective first meiotic division. Under fertilization conditions, the mutant oocytes do not enter into an embryonic development process. Furthermore, we conducted a comparative proteome analysis of wild type and Exosc10 knockout mouse ovaries and identified EXOSC10-dependent proteins involved in many biological processes, such as meiotic cell cycle progression and oocyte maturation. Our results unambiguously demonstrate an essential role for EXOSC10 in oogenesis and may serve as a model for primary ovarian insufficiency in humans.

Project description:This project aimed to elucidate whether human follicular fluid–derived extracellular vesicles (ffEVs) enhance oocyte maturation during rescue in vitro maturation (rIVM). We characterised the protein cargo of ffEVs isolated from follicles containing oocytes at different maturation stages (GV, GVBD, and MII), identifying distinct proteomic signatures associated with follicular competence and oocyte maturity. Subsequently, single-cell proteomic profiling of IVM-matured oocytes supplemented with ffEVs demonstrated that ffEV treatment induces significant changes in oocyte protein abundance, including factors linked to oocyte maturation, endoplasmic reticulum function, steroid biosynthesis, and cytoskeletal organisation. These proteomic alterations were accompanied by changes in oocyte ultrastructure and organelle architecture.
