Expression data from mouse bladder tissues

ABSTRACT: PAI-1 is considered as an oncogenic protein. However, PAI-1 deficiency didn’t stop tumor development in animal cancer models. Thus, it is suggested that there may be compensatory pathway that support tumor growth in the absence of PAI-1. We used microarrays to detail the global gene expression and identified differentially genes between genotypes and treatment.

Project description:The extracellular activity of Plasminogen activator inhibitor-1 (PAI-1) is well described, acting as an inhibitor of tissue plasminogen activator and urokinase-type plasminogen activator, impacting fibrinolysis. Recent studies have revealed a pro-tumorigenic role of PAI-1 in human cancers, via the regulation of angiogenesis and tumor cell survival. In this study, immunohistochemical staining of 939 human bladder cancer specimens showed that PAI-1 expression levels correlated with tumor grade, tumor stage and overall survival. The typical subcellular localization of PAI-1 is cytoplasmic, but in approximately a quarter of the cases, PAI-1 was observed to be localized to both the tumor cell cytoplasm and the nucleus. To investigate the potential function of nuclear PAI-1 in tumor biology we applied chromatin immunoprecipitation (ChIP)-sequencing, gene expression profiling, and rapid immunoprecipitation mass spectrometry to a pair of bladder cancer cell lines. ChIP-sequencing revealed that PAI-1 can bind DNA at distal intergenic regions, suggesting a role as a transcriptional coregulator. The downregulation of PAI-1 in bladder cancer cell lines caused the upregulation of numerous genes, and the integration of ChIP-sequence and RNA-sequence data identified 57 candidate genes subject to PAI-1 regulation. Taken together, the data suggest that nuclear PAI-1 can influence gene expression programs and support malignancy.

Project description:Even after osimertinib is administered to lung adenocarcinomas with EGFR mutations, there are a few cells which survive, and these tolerant cells are considered to be the source of later recurrence. We suspect that PAI-1 is involved in the regulation of these resistant cells and are currently investigating the role of PAI-1 in this process. We used microarray analysis to explore mechanisms involving PAI-1 behind the tolerance to osimertinib in lung adenocarcinoma cells with EGFR mutations.

Transcriptome analysis of normal and cancerous bladder tissues.

Project description:The aim of this research was to explore activation/deactivation of signaling pathways during cancerogenesis. Tissue samples from malignant tumors were obtained from patients who had undergone surgery. Tissue samples from non-cancer controls were collected from autopsies of healthy donors killed in road accidents. Both the tumors and normal tissues were evaluated by a pathologist to confirm the diagnosis and estimate the tumor cell numbers. All tumor samples used in this study contained at least 80% tumor cells. Tissue samples were immediately stabilized in RNAlater (Qiagen) and then stored at M-bM-^HM-^R80M-BM-0C.

Project description:To identify aberrantly expressed long intergenic noncoding RNAs (lincRNAs) in bladder cancer tissues compared with normal adjacent tissues, we have employed microarray expression profiling as a discovery platform to identify lincRNAs that may play important roles in bladder cancer origin and progression. Samples of fresh frozen cancer tissues, together with normal adjacent tissues (3 cm away from the tumor), were obtained during surgical resection, and total RNA was extracted for microarray analysis. Two pairs of fresh frozen bladder cancer tissues and corresponding normal adjacent tissues were used in the microarray analysis.
