LPS-induced and E. coli-induced gene expression in wildtype and MyD88-/- mouse macrophages

ABSTRACT: Comparison of gene expression in wildtype and MyD88-/- C57BL/6J mouse macrophages treated with 10 ng/mL LPS for 2 hours versus media treated control macrophages, and, wildtype and MyD88-/- C57BL/6J mouse macrophages treated with live E. coli bacteria (log phase; 1 bact per 1 macrophage) for 2 hours versus media treated control macrophages. Cells from 4 mice of each geneotype were used and each individual provided its own control. Hybridizations of treated and control samples from each mouse were dye swap replicated. Wildtype macrophages treated with LPS vs control (GSM22617-GSM22623,GSM22625), MyD88-/- macrophages treated with LPS vs control (GSM22626-GSM22632), wldtype macrophages treated with E. coli vs control (GSM22633-GSM22640, and MyD88-/- macrophages treated with E. coli vs control (GSM22641-GSM22648). Project description:Comparison of gene expression in WT and MAL knockout (MALKO) mouse macrophages treated with 10ng/ml lipopolysaccharide (LPS) with that of mock-treated cells incubated for the same time (10 days). Cells from 4 mice of each genotype were used and each individual served as its own control. Hybridizations of treated and control samples were dye swapped. Two experiments: WT vs. LPS-treated WT, and MALKO vs. LPS-treated MALKO. 4 individual biological replicates for each genotype, and each mouse served as its own mock control.

Gene Expression data from mouse bone marrow derived macrophages treated with different inflammatory stimuli

Project description:The activation profiles of macrophages under different immune and inflammatory conditions have generated great interest. LPS, in particular, is a commonly used in vitro model of infection and inflammation studies in macrophages. We have used gene expression microarrays to define the effects of each of three variables; LPS dose, LPS vs. interferons beta and gamma, and genetic background on the transcriptional response of mouse bone marrow-derived macrophages Macrophages derived from the C57BL/6 strain of mouse were challenged with increasing doses of LPS over a 24 hour time-course (0.5ng/ml, 5ng/ml, or 50ng/ml LPS). BALB/c derived macrophages were treated with any of 5ng/ml LPS, 10U/ml of recombinant mouse interferon-beta, or 10U/ml interferon-gamma, over a 24 hour timecourse. Sampling times were 0 hours (pre-treatment) and 1, 2, 4, 8, and 24 hours post-treatment in each case.

Transcription profiling of mouse LPS-stimulated macrophages from wild-type, MyD88-/- and TRIF-/- animals

Project description:Lipopolysaccharide (LPS), a Toll-like receptor (TLR) 4 ligand, activates intracellular signaling via adaptors, MyD88 and TRIF, leading to the expression of various genes including proinflammatory cytokines. We used microarrays to examine influence of MyD88 or TRIF deficiency in LPS-inducible gene expression. Experiment Overall Design: Peritoneal macrophages from wild-type, MyD88-/- and TRIF-/- mice were stimulated with LPS for 0, 1 and 4 hours, followed by RNA extraction. Then hybridization on affymetrix microarrays was performed.

Genome-wide anyalysis of gene expression during prolonged endotoxin tolerance in Aoah -/- mice

Project description:Analysis of gene expression in explanted peritoneal macrophages from Aoah -/- and Aoah +/+ mice treated with LPS 21 days prior to harvest. Explanted peritoneal machrophages were challenged with LPS or control (PBS). The study seeks to characterize global gene expression in the state of prolonged LPS tolerance induced in mice lacking the LPS-inactivating enzyme Aoah. Groups of 9 C57BL/6 Aoah+/+ or Aoah-/- mice were given i.p. injections of 10 µg E. coli LPS/mouse. Twenty-one days later (when Aoah-/- mice remain tolerant and Aoah+/+ mice have recovered), peritoneal macrophages were harvested and the yields from three mice were pooled to form 3 samples per group (i.e., three samples of Aoah+/+ and Aoah-/- mice, with each sample comprised of peritoneal macrophages from 3 mice). Next day, cells were challenged with LPS or PBS and whole RNA was isolated 2 hours later and used for microarray experiments.

ATAC-sequencing of BTBR and C57BL6/J BMDM with Repeated LPS Treatment

Project description:The ability of peripheral innate immune cells to control pro-inflammatory cytokines during periods of repeated or prolonged exposure to immune activating stimuli is critical for preventing tissue damage. In macrophages, repeated exposure to lipopolysaccharide (LPS) induces a refractory period of immune activation termed endotoxin tolerance. Macrophages from the BTBR mouse strain have a skewed pro-inflammatory phenotype and show a hyper-response to LPS challenge. To identify disrupted regulatory mechanisms governing control of cytokine expression in BTBR compared to C57BL/6J mice (C57), we assessed bone marrow derived macrophages (BMDM) treated with either media, one dose or two doses of LPS. Differences in chromatin accessibility using ATAC-sequencing were used to identify regions of differential chromatin accessibility both under baseline media conditions and in response to LPS.

RNAseq analysis of the effects of inorganic long-chain polyphosphates on murine macrophages

Project description:We used unbiased RNAseq analysis from bone marrow-derived macrophages of five individually treated sets of cells from C57BL/6J mice. Cells were treated for 12h with LPS, LPS+L-polyP, L-polyP, or remained untreated. We found several hundred genes to be regulated by the addition of L-polyP. In the combinated treatment with LPS as compared to LPS alone, we found especially interferon-regulated genes to be differentially expressed.
