Heat-shock molecular chaperone Hsp90 is involved in heterochromatin assembly in Schizosaccharomyces pombe.

ABSTRACT: Here, we report the high-throughput profiling of histone modification (H3K9me2) in fission yeast Schizosaccharomyces pombe. We generated genome-wide H3K9me2 maps of fission yeast mutants in swo1-26 (temperature sensitive, ts) cells at 25℃ and 37℃. We find that H3K9me2 enrichment at heterochromatin regions, especially at the mating-type locus and subtelomeres, is compromised, suggesting heterochromatin assembly defects.

The histone chaperone FACT facilitates heterochromatin spreading through regulation of histone turnover and H3K9 methylation states

Project description:Heterochromatin formation requires several steps involving: nucleation followed by spreading of large, silent domains and its faithful re-establishment and maintenance after each replication cycle. The essential histone chaperone FACT has been implicated in heterochromatin silencing, however, little is known about the specificity of FACT in this process. Here, by applying single cell assays, ChIP and genetics, we show that FACT mutants have defects in the spreading of heterochromatin at subtelomeres and mating type locus in fission yeast. Further, we demonstrate that the transition of H3K9me2 to me3 is impaired in the FACT mutant and we link the spreading function of FACT to its role in histone turnover suppression. We also identify Epe1 as a specific factor that counteracts heterochromatin spreading defects in the FACT mutants. Together, our studies identify FACT as a first histone chaperone that promotes heterochromatin spreading and add to the current models on histone turnover in propagation of epigenetic marks.

Histone modification (H3, H3K9ac, H3K9me2) ChIP-seq in WT and cbf11KO strains of Schizosaccharomyces pombe

Project description:Acetylation and dimethylation of lysine 9 on histone H3 are prominent marks of euchromatin and heterochromatin, respectively. Moreover, histone acetylation has been linked to lipid metabolism. We have previously identified the transcription factor Cbf11 as a regulator of lipid metabolism and genome integrity in the fission yeast. Cut6, the acetyl-CoA carboxylase, is one of direct targets of Cbf11. To link the role of Cbf11 in lipid metabolism and chromatin regulation we have performed ChIP-seq of H3K9ac, H3K9me2 and H3 in WT and cbf11KO strains of Schizosaccharomyces pombe in three biological replicates. Strain genotypes: wild type JB32 (h+s); cbf11 knock-out MP44 (h+ cbf11::natR).
