An RNA-Seq atlas of gene expression in rose flower under treatment of various phytohormones and plant growth regulators

ABSTRACT: Using RNA-seq, we recently investigated the transcriptomic dynamics of rose flower under treatment of various plant hormones, including ethylene, 2,4-D, NAA, cytok, gibberellins, abscisic acid, brassinosteroids, salicylic acid, jasmonates, as well as ethylene inhibitor 1-MCP and AgNO. We obtained approximately 240GB data and dissected the transcriptional network with the aim of exploring the transcriptional variation of rose responses towards those plant hormones. Our data will be useful to all those working with the analysis of rose gene expression.

Ethylene and auxin interactions in the roots of Arabidopsis seedlings

Project description:Understanding how developmental and environmental signals are integrated to produce specific responses is one of the main challenges of modern biology. Hormones and, most importantly, interactions between different hormones serve as crucial regulators of plant growth and development, playing central roles in the coordination of internal developmental processes with the environment. Herein, a combination of physiological, genetic, cellular, and whole-genome expression profiling approaches has been employed to investigate the mechanisms of interaction between two key plant hormones, ethylene and auxin. Quantification of the morphological effects of ethylene and auxin in a variety of mutant backgrounds indicates that auxin biosynthesis, transport, signaling and response are required for the ethylene-induced growth inhibition in roots but not in hypocotyls. Analysis of the activation of early auxin and ethylene responses at cellular level, as well as of global changes in gene expression in the wild type versus auxin and ethylene mutants, suggests a simple mechanistic model for the interaction between these two hormones in roots. This model not only implies existence of several levels of interaction but also provides a likely explanation for the strong ethylene response defects observed in auxin mutants. Microarray analysis of cross-talks between Jasmonate acid and ethylene signaling in Arabidopsis seedlings

Project description:Jasmonate and ethylene are two important plant hormones contributing plant resistance to biotic stresses synergistically. Our experimental results showed that two ethylene activated transcription factors (EIN3/EIL1) integrated both jasmonate and ethylene signaling in multiple developmental and defense events. To understand the importance of EIN3/EIL1 in jasmonate signaling, we utilized microarray analysis to identify how much the jasmonate regulated genes are governed by EIN3/EIL1. Our results indicated that EIN3/EIL1 act sequentially in a cascade of transcriptional regulation initiated by jasmonate.

Proteomic analyses of brassinosteroids mutants in response to salt stress in Arabidopsis

Project description:Salt stress is a major environmental factor limiting plant growth and development. Previous studies have indicated that the steroidal hormones—brassinosteroids (BRs) are important regulators of plant responses to salt stress. We found that bes1-D, a gain-of-function mutant of the BR-regulated transcriptional factor BES1 (AT1G19350) in Arabidopsis showed better salt tolerance than the wild-type plant (Col-0). Label-free based LC-MS/MS and proteomic analyses were performed to investigate protein expression in bes1-D and wild-type in response to salt stress.
