Project description:Diffuse intimal thickening (DIT) is a preclinical stage of atherogenesis in humans that does not exist in commonly used mouse models of atherosclerosis. Vascular smooth muscle cells (SMCs) are the predominant cell type that occupies the intimal and medial layers of human coronary arteries. The intimal layer of DIT is particularly prone to developing atherosclerosis later in life. Differences in the molecular features between the intimal and medial layers of DIT regions may represent the earliest changes that promote atherogenesis in humans, and can be studied by spatial molecular phenotyping.

Project description:Purpose: To investigate the effects of high cholesterol diet on the intimal transcriptome related to atherosclerosis. Methods: 3-week-old LDLR-/- male mice were fed a high cholesterol or low cholesterol diet for 9 weeks. Intimal RNA was extracted by injecting Trizol into the aorta and collecting the flow through for further processing. Results: Mice fed a high cholesterol revealed upregulation in atherosclerosis and immune/inflammatory related genes.

Project description:Abdominal aortic aneurysm (AAA) is an asymptomatic chronic disease of the aorta and its evolution is unpredictable. Despite the existence of a number of pathological mechanisms contributing to the dilation of the human AAA wall, there is currently no specific therapy to prevent the fatal rupture of the aorta. Our objective was to identify novel mediators and/or biomarkers involved in the instability of the aortic wall that could help to prevent AAA progression.

Effects of paternal high cholesterol diet on F1 generation intimal transcriptome

Project description:Purpose: To investigate the effects of paternal high cholesterol diet on the F1 intimal transcriptome related to atherosclerosis. Methods:Three-week-old male LDL Receptor-deficient (LDLR-/-) male mice were fed a low-cholesterol diet (LCD) or high-cholesterol diet (HCD) for 8 weeks and then mated with age-matched control LDLR-/- female mice to generate F1 offspring. F1 LDLR-/- mice were fed a LCD for 16 weeks. Intimal RNA was extracted by injecting Trizol into the aorta and collecting the flow through for further processing. Results: . Total RNA isolated from the F1 intima were then subjected for RNA-sequencing analysis and results show that paternal hypercholesterolemia led to upregulation of 147 genes in the intimal of F1 female mice, many of which are involved in immune and inflammatory pathways.

Bulk RNA-seq of foamy and non-foamy intimal macrophages from ApoE -/- mouse

Project description:Macrophages in atherosclerotic aorta are major population in lesion and contributes to lesion formation by becoming foam cells. To investigate in vivo transcriptome profiles of those macrophages, we extracted foamy and non-foamy macrophages from atherosclerotic aorta using lipid probe-based flow cytometry sorting. Our data indicates that intima non-foamy and foamy macrophages show different mRNA expressions. Rather than non-foamy macrophages, foamy macrophages expressed more genes related to cholesterol metabolism, oxidative phosphorylation, lysosome and so on. The non-foamy macrophages expressed more genes related to immune response (Il-1b related pathways, TNF, TLR signaling pathways) than foamy macrophages.

CD10 expression in human adventitial cells

Project description:The tunica adventitia ensheaths arteries and veins and contains presumptive mesenchymal stem cells (MSCs) involved in vascular remodeling. We show here that a subset of human adventitial cells expresses the CD10/CALLA cell surface metalloprotease. Sorted CD10+ adventitial cells developed promptly in culture into MSCs, exhibiting higher proliferation, clonogenic and osteogenic potentials than CD10- counterparts. CD10+ adventitial cells increased expression of the cell cycle protein CCND2 via ERK1/2 and prompted NELL1 expression via NF-κB. CD10 expression was upregulated in CD10+ adventitial cells through neural secreted protein sonic hedgehog-mediated Gli1 signaling. These results suggest that CD10 expression, which marks rapidly dividing cells in other normal and malignant cell lineages, plays a role in MSC homeostasis in human tissues.
