Project description:Gene expression of rat peripheral blood mononuclear cells was analyzed by microarray analysis in normoweight and in diet-induced obese rats (cafeteria rats). The aim of this study was to identify genes involved in energy homeostasis that are altered in the obese state. Keywords: Dietary treatment, obese-state analysis This study analyzed the gene expression of PBMC in normoweight and in diet-induced obese (cafeteria-fed) Wistar rats submitted to ad libitum feed conditions. Two-month-old male Wistar rats (n=10) were assigned into two dietary groups for 4 months: the control group (n=5) was fed with a standard chow diet, whereas the second group (cafeteria group, n=5) was fed with a fat-rich hypercaloric cafeteria diet in addition to the standard chow. At 6 months of age, blood samples were collected in feeding conditions from the safena vein. Peripheral blood mononuclear cells were isolated by Ficoll gradient separation, and RNA was extracted. One sample in the control group was excluded because of a low amount of RNA. Gene expression changes were assessed using an Agilent rat whole genome microarray (G4131F Agilent Technologies).

Project description:In a randomized controlled dietary intervention study we compared an isocaloric Healthy Nordic diet with the average Nordic diet for influence on peripheral blood mononuclear cells (PBMC) gene expression. We studied obese adults with features of the metabolic syndrom, n=66. There was no significant difference in age, BMI, or gene expression between the groups before the intervention. The intervention lasted for 18-24 weeks. Blood samples were obtained before (W0), during (W12) and after completion (W18-24) of the dietary intervention. From the samples we extracted RNA. From total RNA we prepared and hybridised biotinylated complementary RNA to GeneChip Human Gene 1.1 ST Arrays (Affymetrix Inc., Santa Clara, CA), and then washed, stained and scanned the slides using standardised protocols (Affymetrix Inc.).

The role of dietary leucine and dairy protein on hepatic gene expression

Project description:We used microarray to determine the differences in hepatic gene expression for diet-induced obese Sprague-Dawley rats consuming different dietary proteins. Proteins of interest included skim milk powder (dairy), casein, and the branched-chain amino acid, leucine. The primary aims of this study were: (i) to compare the effects of diets with protein derived from casein, casein supplemented with leucine, and complete dairy on body composition and insulin sensitivity; and (ii) to determine if there is a synergistic effect of dietary Ca and protein source on body composition and insulin sensitivity. Secondarily, we used microarray analysis to examine the effect of casein, leucine, or complete dairy containing diets on the expression of hepatic genes related to lipid and glucose metabolism. Diet induced obese rats consumed ad libitum, a high fat, high sucrose diet for 8 weeks (n=4). All diets had an energy density of 4.6 kcal/gram and provided 10% of total energy from protein [casein, complete dairy (skim milk powder), or leucine-supplemented casein (7.1% from casein plus 2.9% from leucine)]. The casein treatment was the control diet.

Comparison of radiation-induced rat mammary carcinomas from individuals with different sensitivity to diet-induced obesity

Project description:Understanding the relationship between radiation-induced breast cancer and obesity, together with information on underlying mechanisms, are potentially useful in risk management and prevention of second cancer in patients receiving radiotherapy. The present study aims to develop a novel animal model to study the relationship by combining two Sprague-Dawley rat models of radiation carcinogenesis and diet-induced obesity. Mammary carcinomas were induced in female obese and lean rats by irradiation with 4 Gy of gamma rays. Gene expression of mammary carcinomas and normal mammary tissues were analyzed with Agilent Whole Rat Genome DNA microarray. The result indicated that genes related to translation and oxidative phosphorylation were upregulated in carcinomas of obese rats.

Project description:The aim of this study was to analyze gene response to a 10-week dietary intervention for weight loss in peripheral blood mononuclear cells of overweight/obese male children. PBMCs were obtained from 12 overweight/obese boys for RNA extraction and hybridization on Affymetrix microarrays. We performed the microarray analysis at baseline and after a weight loss intervention program in a total of 24 samples. They were distributed by dietary response as high and low responders.
