Neutrophil modulation by IL-1β-induced fibroblasts

Project description:Neutrophils are key contributors to the pathogenesis of SLE. Using single-cell sequencing data from SLE skin lesions, we identified a subcluster of CXCL1+ fibroblasts primarily induced by IL-1β. To better understand the effects of CXCL1+ fibroblasts on neutrophils, we established a conditional culture model using culture supernatants from control and IL-1β-stimulated fibroblasts. Neutrophils were collected after 21 hours of conditional culture and subjected to RNA-seq. Our findings revealed that neutrophils exhibited a distinct pro-inflammatory phenotype, expressing higher levels of pro-inflammatory cytokines such as IL1B, TNFA, IL8, and OSM.

Platelets direct monocytes differentiation into multinucleated giant foam cells with suppressive response to mycobacterial stimulus

Project description:Co-culture of platelets with monocytes induces the forming of multinucleated giant foam cells (MP-Mac or MP-MNGCs). Compared with normal monocyte differentiated macrophages (M-Mac), the MP-MNGCs can engulf more mycobacteria with suppressed inflammatory responses (releasing more IL-10 with less TNF-gamma). Microarray analysis demonstrated that the MP-MNGCs greatly up-regulated CXCL5, PPBP, NT5E, PDPN, CXCL3 and other M2 related genes, such as ARG1, CXCL1, CD163, etc. GO analysis revealed higher cluster of genes in response to wounding and nuclear division. Immunohistochemistry studies indicated similar high expression of IL-10, CXCL5, CXCL3, PDPN and ARG1 expression in MNGCs of TB granuloma, which indicated that phagocytosis of activated platelets by monocytes might be involved in pathogenesis of TB granuloma. Microarray experiments were performed as dual-color hybridizations. To compensate for dye-specific effects, a dye-reversal color-swap was applied. Quality control and quantification of total RNA amount was assessed using an Agilent 2100 bioanalyzer (Agilent Technologies) and a NanoDrop 1000 spectrophotometer (Kisker).

Transcriptional profiling of long-term stimulated primary murine intestinal fibroblasts with interleukin-36.

Project description:Previous stimulation experiments of stimulated primary murine colonic fibroblasts with IL-36R ligands for 4h in vitro showed an upregulation of pro-inflammatory cytokines e.g. IL-6, IL-1b and chemokines e.g. CXCL1, CCL2 indicating an important role for IL-36 in inflammation. We were interested in analyzing long-term stimulation (9 days) of intestinal fibroblasts to identify a putative differential expression profile. The experimental setup included 6 samples from 3 colons in a pairwise design (3 stimulated vs 3 unstimulated).

Project description:Multicellular cytokine networks regulate the onset of intestinal inflammation and colitis-associated cancer (CAC). Interleukin 22 (IL-22) promotes epithelial cell recovery but can also drive inflammation and tumorigenesis. We demonstrate that IL-22 from innate lymphoid cells activates STAT3 and increases OSM receptor expression in intestinal epithelial cells. This activation leads to sustained STAT3 activity via OSM, promoting inflammation and tumorigenesis. Deleting the OSM receptor or blocking OSM pharmacologically mitigates colitis and CAC. Our findings highlight the IL-22-OSM axis as a potential therapeutic target for these conditions.

Project description:Multicellular cytokine networks orchestrate the onset and progression of intestinal inflammation and colitis-associated cancer (CAC). Interleukin 22 (IL-22), a member of the IL-10 superfamily, is known for promoting epithelial cell recovery but can inadvertently fuel inflammation and tumorigenesis. Here, we demonstrate that IL-22, derived from group 3 innate lymphoid cells (ILC3), triggers oncostatin M (OSM) responsiveness in intestinal epithelial cells by activating STAT3 and upregulating OSM receptor (OSMR) expression. OSM, a member of the IL-6 cytokine family implicated in inflammatory bowel disease, collaborates with IL-22 to sustain STAT3 activation, promoting proinflammatory adaptations and immune cell chemotaxis to the intestine. Conditional deletion of OSMR in epithelial cells protects mice from colitis and CAC. Additionally, pharmacological blockade of OSM reduces the progression of established CAC. Our study reveals a novel mechanism by which OSM sustains intestinal inflammation and CAC, identifying the IL-22-OSM axis as a promising therapeutic target.
