Genome wide RNA-sequencing of human pluripotent stem-cell derived endothelial cells 48h after transduction with adenoviruses expressing either empty vector of different combination of transcription factors

ABSTRACT: To induce barrier of stem-cell derived endothelial cells in vitro cells have been transduced with a mixture of adenovirus overexpressing transcription factors with potential role in endothelial cell barrier stabilization. Adenoviruses overexpressing ETS1, SOX18 and SOX7 have been part of both transcription factor mixtures. As fourth transcription factor in the mixture included TAL1 while the other mixture included LEF1. Each transcription factor was used at 20 MOI. As control adenovirus overexpressing empty vectors has been used (80 MOI).

ChIP-seq for E4BP4 in CD4+ T cell

Project description:Here, using ChIP-seq, we demonstrate that the transcriptional repressor Adenovirus E4 promoter-binding protein (E4BP4) binds directly to the Bcl6 promoter, which a key transcription factor controlling Tfh cell differentiation. By obtaining sequence from chromatin immunoprecipitated DNA of E4BP4 overexpressing CD4+T cells, we generated genome-wide binding gene spectrums of E4BP4. These results reveal that E4BP4 interacts with BCL6 and E4BP4 directly modulated the expression of Bcl6 to reveal the mechanism downstream of E4BP4 that regulates Tfh cell differentiation.

Transcriptome Data of SREBP2(N) Overexpression in Human Umbilical Vein Endothelial Cells

Project description:Sterol regulatory element-binding protein 2 (SREBP2) is a transcription factor that has been recently discovered to mediate vascular endothelial cell (EC) dysfunction. We therefore investigated the role of SREBP2 in EC function through comparing the transciptional profiling of human umbilical vein endothelial cells (HUVECs) that were infected with adenovirus overexpressing SREBP2 (Ad-SREBP2) or with empty vector (Ad-null) control. Gene ontology analysis revealed that SREBP2 not only regulates cholesterol biosynthesis, but also mediates the TGF, WNT, and cytoskeleton remodeling pathways. Among the responsive genes, SREBP2 exhibited the induction of mesenchymal transition.

Quantitative analysis of tet inducible SOX18 during hematopoietic differentiation and Natural killer cell differentiation

Project description:Using inducible SOX18 human PSCs, we found overexpression of SOX18 showed change of transcriptional program at D8 especially T cell receptor and Toll like receptor signaling pathways. So this suggests SOX18 has an effect on lymphoid differentiation program. Specifically overexpressed SOX18 enhance NK cell production, and related to metabolism pathway. This finding that SOX18 promotes development of progenitors with superior NK cell potenital will advance our understanding of molecualr network regulating specification on lymphoid cells and can help to improve the scalability of NK CELL production form hPSCs form immunotherapies.
