Drosophila melanogaster transcriptional profiling over life span: Indy vs. Controls

Project description:Transcriptional profiling of Indy long lived flies and controls over the course of their entire lifespan. Mutations in the Indy gene extend life span in Drosophila melanogaster. This study investigates the changes in gene expression over time in Indy206 flies heterozygous over Canton-S (Indy206/CS) and compares them to genetically matched heterozygous controls (2216/CS). Samples from both fly strains were collected at age: 5, 10, 20, 30, 40, 50, 70 and 80.

Project description:Timecourse experiment (0hr, 2hr,6hr and 21hr) where RNA was extracted from non-rested PBMCs which were exposed to 4 different conditions- nothing (mock), interferon (IFN)gamma, IFN gamma and a non-specific antibody and IFN gamma with purified patient antibody. The purified patient antibody is demonstrated to have a specific anti-interferon gamma effect over time, with the expression profile in the arrays with IFN gamma plus patient antibody most closely resembling the mock arrays. Time series experiment on L4 stage of Caenorhabditis elegans N2

Project description:This experiment investigates the transcriptional effect of age in the N2 strain of Caenorhabditis elegans during the L4 stage. The goal is to identify genes that show dynamic gene-expression patterns over time. We grew N2 populations over the course of 44 to 58 hours at 20 degrees Celsius, independent populations were harvested in hourly intervals. Thereafter RNA was isolated, labelled and hybridized on microarray. The gene expression profiles were analyzed for dynamic expression patterns using principal component analysis and k-means clustering.
