Project description:The purpose is to obtain samples for mRNA analysis in human U937 cells infected with Zaire Ebola virus wild-type in the deltaVP30 background and delta-mucin virus. Human U937 cells (monocyte-like) expressing the Ebola VP30 protein were infected with Zaire Ebola virus wild-type (wild) in the delta-VP30 background and delta-mucin virus (encodes a GP lacking the mucin domain) (mucin). Infected samples were collected in quintuplet; time-matched mocks were collected in quintuplet in parallel with infected samples. Time points: 0, 8, 18, and 30 h post-infection.

Genome-wide localization of Swi6 and H3-Lys9-me2 in wt and mutant starins of S. pombe

Project description:ChIP-on-ChIP experiments were performed to quantitate the genome-wide localization of heterochromatin proteins Swi6 and H3-Lys9-me2 in wild type versus polalpha mutant swi7H4, Pol delta mutant, dts2 and cut4 mutant cut4-993. Our results indicate that the localization of Swi6 and H3-Lys7-me2 at heterochromaitn regions, like centromere, telomere and mating-type is abrogated in the swi7H4, dts2 and cut4 mutants.

Project description:Methylation of CpG islands is associated with transcriptional repression and, in cancer, leads to the abnormal silencing of tumor-suppressor genes. Genome wide methylation profiling of myeloid leukemia cell lines identified a large number of genes with aberrantly methylated CpG islands. Comparative mRNA expression analysis suggests that more than half of these genes show extremely low or absent expression in normal cells, suggesting that hypermethylation in cancer may be independent of the transcriptional status of the affected gene. Experiment Overall Design: The expression profiles of the three leukemia cell lines KG-1, U937 and THP-1 was compared to normal human blood monocytes (reference). The set includes a single hybridization for each sample.

Project description:H3K27Ac is one of the expressed enhancer markers, PPARβ/δ is a transcription factor and Pol II (RNA polymerase II) is an enzyme which catalyzes the transcription of DNA to synthesize precursors of mRNA and most snRNA and microRNA. These genomic localization in endothelial cells is unknown in endothelial cells. This time, we established a new antibody for H3K27ac, PPARβ/δ and Pol II and performed ChIP-seq to identify H3K27ac, PPARβ/δ and Pol II binding site in whole genome manner under PPARβ/δ agonist and/or hypoxia.
