PIM kinase inhibition in HCC xenograft

ABSTRACT: Human HCC cell line, MHCC-97L, was subcutaneously injected to BALB/c nude mice. Two weeks post injection, mice were randomised to receive vehicle or PIM447 (60mg/kg) by oral gavage three times a week for two weeks. Total RNA was extracted from the tumor xenograft (Vehicle, n=2; PIM447, n=2) and subjected to RNA sequencing. Expression profiling of the tumors between vehicle control and PIM447 treated groups was compared.

Hepatic transcriptome analysis after acute and chronic glucocorticoid exposure combined with androgen receptor antagonism.

Project description:Aim: We investigated the effect of acute and chronic glucocorticoid exposure on the hepatic transcriptome and its dependency on active androgen signaling. Methods: Adult male mice were used in the experiments. For the chronic exposure, mice were implanted with CORT (20mg) or vehicle pellets and administered ENZA (40mg/kg/day) or solvent via oral gavage. For the acute exposure, mice were pre-treated with ENZA (40mg/kg/day) or solvent every 12 hours for 24 hours via oral gavage and subsequently subcutaneously injected with 3mg/kg CORT or vehicle. Livers of all animals were dissected after 3 hours and snap-frozen. Next, total RNA was isolated and send for 100bp paired-end sequencing by BGI.

Analysis of changes in transcriptomes of the rat medial prefrontal cortex tissues (mPFC) in vehicle group, chronic mild stress (CMS) group, and CMS+Phloretin group

Project description:Purpose: The goals of this study are to investigate the effects of Phloretin (PHL) on the changes of genes in rat mPFC tissues between Vehicle group, CMS group, and CMS+PHL group. Methods: Vehicle group: Rats received vehicle by oral gavage for 2 weeks, and then were left undisturbed for 3 weeks except for necessary procedures, including handling and routine cleaning for 3 weeks. CMS group: Rats received vehicle by oral gavage for 2 weeks, and then were subjected to eight different stressors for 3 weeks in a random order, including foot shock, water deprivation, cold water swimming, food deprivation, restraint, clip tail, light/dark cycle reversal, and cage tilt. CMS+PHL group: Rats were pre-treated with 20 mg/kg. Phloretin for 2 weeks, and then exposed to above eight stressors randomly together with 20 mg/kg Phloretin for 3 weeks. In each group, we collected mPFC tissues from 3 rats and total RNA was extracted. RNA integrity was assessed using the RNA Nano 6000 Assay Kit of the Bioanalyzer 2100 system. The clustering of the index-coded samples was performed on a cBot Cluster Generation System using TruSeq PE Cluster Kit v3-cBot-HS (Illumina) according to the manufacturer’s instructions. After cluster generation, the library preparations were sequenced on an Illumina Novaseq platform and 150 bp paired-end reads were generated. Results: We found that Phloretin treatment changed some gene expression, especially in neuronal plasticity, compared with CMS group.

Microarray analysis of rat testis following combined fetal and postnatal DBP exposure

Project description:Provided later Pregnant Fisher 344 rats will be purchased from Charles River Laboratories, Inc. and delivered to CIIT on gestational day (GD) 7 (GD0 = day first vaginal plug positive). At gestational day 12 (GD12), the dams will be exposed once/day until GD20 to 50 mg/kg dibutyl phthalate (DBP) in corn oil vehicle via oral gavage. Each dose group will contain 4-6 vehicle control or phthalate treated dams. Groups of animals will be sacrificed at GD20, postnatal day (PND) 35, and PND90 for endpoint analysis. At GD20, treated and control animals will be examined for various endpoints including body weight, testicular histopathology, gene expression profile via microarray analysis, and anogenital distance (AGD). AGD (at parturition; PND1) and nipple number/location (at PND14 and day of sacrifice) will be determined on animals in the postnatal groups. At PND35 or 90, one male from each in utero corn oil vehicle or DBP exposed group will receive a second gavage of either corn oil or 500 mg/kg DBP. 6 hours after the second gavage, the following endpoints will be examined: 1) testis histopathology; 2) spermatid head quantification (PND90 only); 3) testis and body weights; 5) genome-wide gene expression (via microarray); and 6) germ cell apoptosis (TUNEL assay).

The effect of GPR30 agonist G-1 on androgen-sensitive and castration-resistant LNCaP xenografts

Project description:G-1 is an agonist to GPR30. Activation of GPR30 by G-1 inhibited prostate cancer cell growth in LNCaP xenografts regrown after catration of the host (nude mice), but not in the androgen-sensitive LNCaP xenograft grown in an intact host. Results provide insights into the molecular basis of G-1 action in castration-resistant prostate cancer. Male nude mice were injected with LNCaP cells. When the LNCaP tumors reached 150–300 mm3, mice were divided into two groups: intact (androgen-sensitive tumor) and castrated. For the intact group, mice were subcutaneously injected with vehicle alone (95% PBS, 2.5% DMSO, 2.5% ethanol) or G-1 (4 mg/kg/day in vehicle) daily for 16 days. For the castrated group, tumors regressed and then regrew to ~300-400mm3. Mice were treated daily with vehicle or G-1 as described for 16 days. Tumors were harvested for RNA extraction and microarray experiments.

The anti-fibrosis effect of iguratimod on skin tissues of systemic sclerosis patients

Project description:To evaluate the possibility of iguratimod working on SSc patients in clinical practice, we collected skin biopsies from three early SSc patients. Three grafts were divided equally into two irradiated nude mice and treated with iguratimod or vehicle solution subcutaneously for five weeks. Total RNA from cultured fibroblasts or xenografts were extracted by TRIzol.Differential expression analysis of two groups was performed using the DESeq2 from R package. we find the ensemble of genes encoding extracellular matrix and ECM-associated proteins, which was termed as matrisome, was enriched at the top of the GSEA list.In the ensemble, there were pro-fibrotic cytokine genes such as TGFB1 and FGFs, and WNTs; ECM degradation enzymes genes such MMPs and CTSs; ECM-associated chemokine such CCL13 and CCL18; and notably, remarkable amount of glycoprotein/ proteoglycan genes such as LRG1, PRG4, HAPLN1 and PODNL1.

Effects of oxysterol Oxy186 and Oxy210 on A549 xenograft tumor growth

Project description:To examine the efficacy of oxysterols in inhibiting tumor growth in vivo, we tested Oxy186 and Oxy210 in subcutaneous xenograft tumor models constructed with A549 cells in nude mice. After the tumor size reached an average of ~100 mm3, the compounds were administered daily via oral gavage at 50 mg/kg. We found that Oxy186 started to inhibit tumor growth at day 8, and significantly reduced the tumor volume from day 11 to day18 of treatment, compared to that of vehicle control. Surprisingly, Oxy210 did not show such inhibitory effect.
