The experiment, compared transcriptional profiles from SP and MP (Main Population) cells from either C2C12 or FGF6-expressing C2C12.

UB/OC-1 differentiation

Project description:This study explores the profiles of genes expressed during conditional differentiation of inner ear hair cell precursors in vitro. The cell line UB/OC-1 was derived from mouse embryonic organs of Corti (Rivolta et al, Proc. R. Soc. Lond. B 265: 1595-1603, 1998) and it can be induced to differentiate upon a temperature shift. The study has been replicated in two separate populations of cells, named Exp A and Exp B. Exp A includes daily time points from 0 to 9 days, then 11 and 14 days. Exp B includes days 0, 2, 4, and 9. The experiment has been repeated 4 times (exp IFX1-IFX3-CPLA2-CPLA3)and samples were harvested around 11h in the morning (ie 3h after illumation).

Project description:Transcriptome analysis of Casz1 was performed using litters of postnatal day 2 retinas from crosses between Casz1Flox/Flox; R26-Stop-EYFP and Casz1Flox/Flox; IKCre-A; R26-Stop-EYFP. The IK-A Cre driver is described in Tarchini et al., Dev Dyn 241(12):1973-85. EYFP marks Cre expressing (and therefore cKO) cells. Cells were incubated for 30 min in RGM serum-free medium described in Cayouette et al., Neuron 40(5):897, supplemented with Hoechst 33342 (Invitrogen), and 4N cells were sorted using a MOFLO cytometer (Beckman) based on Hoechst, GFP expression, and propidium iodide exclusion.

Project description:The vacuole occupies a large portion of plant cell volume, it is especially true to fruit tissues. Berry flesh cell vacuole serves as storage organelle for water, sugars, acids, secondary metabolites and others, which largely determining berry quality (Fontes et al., 2011a, b; Shiratake and Martinoia, 2007, Conde et al., 2006). However, the molecular basis of these compartmentation processes is still poorly understood. As in many species, the major bottle neck to study these aspects in grapevine is to obtain highly purified vacuoles with a good yield (Fontes et al., 2010). Up to date, several vacuole or tonoplast proteome researches were applied on a few plants mainly on Arabidopsis thaliana, vacuoles or tonoplast were derived from mesophyll cells (Carter et al., 2004, Endler et al., 2006, Schulze, et al., 2012) or cell culture (Jaquinod et al 2006, Shimaoka et al 2004), cauliflower buds (Schmidt et al., 2007) and sugar beet taproots (Jung et al., 2015). Though the grape berry protoplasts and intact vacuoles were successfully isolated from Cabernet Sauvignon berry suspension-cultured cells (Fontes et al., 2010), the vacuoles isolated from grape berry or different development and ripening stages of grape berry mesocarp tissues were not achieved.

Project description:Recent developments in animal models (Morris et al., 2004; Tumbar et al., 2004) as well as the discovery of cell surface markers (Jones and Watt, 1993; Tani et al., 2000; Trempus et al., 2003; Nijhof et al., 2006) have made it possible to isolate living epidermal hair follicle stem cells (HFSCs) from mouse skin, facilitating the study of the biological and molecular features inherent to HFSCs. A complexity of stem and progenitor cell populations within the hair follicle has been revealed. Here, we report comprehensive profiling of mouse CD34-expressing HFSCs using the Agilent mouse oligo microarray platform in order to extend and enrich the existing HFSC databases.
