Profiling anagen skin in control and Fgf5 mutant mice

ABSTRACT: We report transcriptomes of cells from adult mouse skin in anagen phase of the hair growth cycle, comparing control tamoxifen induced Nfatc1-CreER;mTmG mice with mutant tamoxifen induced Nfatc1-CreER;mTmG;Fgf5-/- mice. Dead cells were removed from whole skin tissue using dead cell removal kit (MACS). Remaining live cells were analyzed.

Bmal1 controls circadian cell proliferation and susceptibility to UVB-induced DNA damage in the epidermis [Anagen]

Project description:While several physiological skin parameters vary in a circadian manner, the identity of genes participating in chronobiology of skin remains unknown, leading us to define the circadian transcriptome of mouse skin at two different stages of the hair cycle, telogen and anagen. The circadian transcriptomes of telogen and anagen skin are largely distinct, with the former dominated by genes involved in cell proliferation and metabolism. The expression of many metabolic genes is antiphasic to cell cycle related genes, the former peaking during the day and the latter peaking at the night. Consistently, accumulation of reactive oxygen species, a byproduct of oxidative phosphorylation, and S-phase are antiphasic to each other in telogen skin. Furthermore, the circadian variation in S-phase is controlled by BMAL1 intrinsic to keratinocytes as keratinocyte-specific deletion of Bmal1 obliterates time of day dependent synchronicity of cell division in the epidermis leading to a constitutively elevated cell proliferation. Consistent with higher cellular susceptibility to UV-induced DNA damage during S phase, we found that mice are most sensitive to UVB-induced DNA damage in the epidermis at night. As maximum numbers of keratinocytes go through S phase in the late afternoon in the human epidermis, we speculate that in humans the circadian clock imposes regulation of epidermal cell proliferation such that skin is at a particularly vulnerable stage during times of maximum UV exposure, thus contributing to the high incidence of human skin cancers. Whole skin was collected at 4 hour intervals for 48 hours. ZT number indicates the number of hours elapsed from when lights are switched on. ZT0 = lights on (6am). ZT12=lights off (6pm). Total RNA was purified from the skin of each mouse and equal amount of RNA from the 3 replicates for each time point were pooled. Anagen samples were collected from skin of P30 male mice.

Comparative Proteomic Analysis of the Telogen-to-Anagen Transition in Cashmere Goat Secondary Hair Follicles

Project description:This study investigates the proteomic changes associated with the telogen-to-anagen transition in secondary hair follicles (SHFs) of cashmere goats. Using tandem mass tag (TMT) labeling combined with LC-MS/MS, we identified 3,654 proteins, with 458 differentially expressed proteins (DEPs) between telogen and anagen phases. Key proteins such as ADAM17, SFRP1, and PPP1CA were found to play crucial roles in regulating SHF cycle transitions. These findings provide insights into the molecular mechanisms underlying cashmere fiber growth and offer potential targets for improving cashmere production.

Next Generation Sequencing Facilitates Quantitative Analysis of Control and beta-catenin Deficient Cancer-associated Fibroblasts Transcriptomes

Project description:D4M BrafV600E; Ptenlox5/lox5 melanoma cells mixed with uninduced control Fb (Ctnnb1+/+; Col1α2-CreER) or mutant bcat/Fb (Ctnnb1fl/fl; Col1α2-CreER) were injected intradermally into flanks of control Ctnnb1+/+; Col1α2-CreER or Ctnnb1 fl/fl; Col1α2-CreER mice, respectively. Tumors were allowed to grow to a volume of approximately 62.5 cubic millimeters when tamoxifen (TAM) was then administrated to induce β-catenin ablation in mutant bcat/Fb CAFs. Control CAFs or bcat/CAFs were collected from BrafV600E; Ptenlox5/lox5 melanoma tumors using PDGFRα as a marker for fibroblasts. Normal fibroblasts were collected from B6 mouse skin.

Identification of genes and proteins associated with anagen wool growth

Project description:Sheep provide considerable materials for the animal fibre industry. Identifying genes of major effect for wool growth would offer strategies for improving the quality and increasing the yield of fine wool. In this study, we employed Agilent Sheep Gene Expression Microarray and proteomic technology to investigate the gene expression patterns of body side skin (more wool growing) in Aohan fine wool sheep (a Chinese indigenous breed) in comparison with groin skin (no wool growing) at the anagen stage of wool follicle. Microarray study revealed that 4772 probes were differentially expressed, including 2071 upregulated and 2701 downregulated probes in the comparisons of body side skin versus groin skin (S/G). The microarray results were verified by means of quantitative PCR. 1099 probes were assigned to unique genes/transcripts. The number of distinct genes/transcripts (annotated) was 926, of which 352 were up-regulated and 574 were down-regulated. In S/G, 13 genes were up-regulated by more than 10-fold, while 60 genes were down-regulated by more than 10-fold. Further analysis revealed that the majority of the genes possibly related to the wool growth could be assigned into the categories including regulation of cell division, intermediate filament, cytoskeletal part and growth factor activity. Several potential gene families might participate in hair growth regulation, including fibroblast growth factors, transforming growth factor-β, WNTs, insulin-like growth factor, vascular endothelial growth factors and so on. Proteomic analysis also got 196 differentially expressed protein points, of which 121 were identified as single protein points. Furthermore, according to the results at both mRNA and protein levels, similar regulation mechanism of gene activity might be engaged during skin development and embryo development.

Project description:We isolated and selected intestinal adenoma organoids from villin-CreER; Apcflox/flox and villin-CreER; Apcflox/flox; Prox1flox/flox mice and added tamoxifen to induce the deletion of the Apc and Prox1 genes in the intestinal epitheliul ex vivo. Microarray experiments were carried out 7 days after the addition of tamoxifen. Total RNA obtained from villin-CreER; Apcflox/flox and villin-CreER; Apcflox/flox; Prox1flox/flox organoids were compared 7 days after the addition of tamoxifen and 5 days after the selection for Apc-mutant organoids in the absence of the Wnt-agonist R-Spondin1.
