RNAseq data for murine BMDM infected with wild-type Mycobacterium tuberculosis, a PDIM mutant, or an ESX-1 mutant.

ABSTRACT: C57BL6 murine bone marrow-derived marcrophages were infected with Mycobacterium tuberculosis H37Rv, a PDIM mutant (H37RvΔmas) or an ESX-1 mutant (Tn::eccCa1) strains at an MOI of 2:1. RNA was harvested at 4, 8, 12, and 16 hours post-infection for comprehensive transcriptional profiling.

Secretome and intracellular proteome of espB deletion mutant of Mycobacterium tuberculosis

Project description:Mycobacterium tuberculosis relies on the ESX-1 secretion system for survival, multiplication in and escape from macrophages. In this work we investigate the role of the EspB protein, encoded within the esx-1 gene cluster, in virulence and ESX-1 substrate secretion in M. tuberculosis H37Rv. Genetic, proteomic and immunological data show that, contrary to previous reports, an espB knock-out mutant is only partially attenuated in ex vivo infection models, where EsxA, EsxB and EspC antigen presentation is not affected, and secretes the major virulence factor EsxA. Additionally, we demonstrate that EspB does not require an intact and functional ESX-1 apparatus for being secreted in H37Rv and in M. bovis BCG, as opposed to other strains such as CDC1551 and Erdman, thereby suggesting that other ESX systems may be involved in the process. Overall our findings highlight unexplored differences in the secretion profiles of various mycobacterial strains and underscore the plasticity of ESX-dependent secretion in mycobacteria.

Virulence regulator EspR of Mycobacterium tuberculosis is a nucleoid-associated protein

Project description:The principal virulence determinant of Mycobacterium tuberculosis (Mtb), the ESX-1 protein secretion system, is positively controlled at the transcriptional level by EspR. Depletion of EspR reportedly affects a small number of genes, both positively or negatively, including a key ESX-1 component, the espACD operon. EspR is also thought to be an ESX-1 substrate. Using EspR-specific antibodies in ChIP-Seq experiments (chromatin immunoprecipitation followed by ultra-high throughput DNA sequencing) we show that EspR binds to at least 165 loci on the Mtb genome. Included in the EspR regulon are genes encoding not only EspA, but also EspR itself, the ESX-2 and ESX-5 systems, a host of diverse cell wall functions, such as production of the complex lipid PDIM (phenolthiocerol dimycocerosate) and the PE/PPE cell-surface proteins. EspR binding sites are not restricted to promoter regions and can be clustered. This suggests that rather than functioning as a classical regulatory protein EspR acts globally as a nucleoid-associated protein capable of long-range interactions consistent with a recently established structural model. EspR expression was shown to be growth phase-dependent, peaking in the stationary phase. Overexpression in Mtb strain H37Rv revealed that EspR influences target gene expression both positively or negatively leading to growth arrest. At no stage was EspR secreted into the culture filtrate. Thus, rather than serving as a specific activator of a virulence locus, EspR is a novel nucleoid-associated protein, with both architectural and regulatory roles, that impacts cell wall functions and pathogenesis through multiple genes. Project description:Genomic comparisons among PDIM positive and negative isolates. Standard in vitro exponential phase cultures of M. tuberculosis H37Rv ATCC27294 were plated on 7H11 plates. Individual colonies were sub-cultured in 7H9 broth and metabolically labeled with [14C] propionic acid. The apolar lipid fraction was then extracted from those cultures and assayed for PDIM (phthiocerol dimycocerosate) content via TLC. Individual isolates producing and deficient in PDIM were selected for performing genomic comparisons.
