RNA-seq of mouse suprarenal aorta from VAMP8-knockout and wild-type mice with AngII or saline infusion

Project description:VAMP8 (vesicle-associated membrane protein 8) is a SNARE protein implicated in exocytotic secretion from immune and vascular cells. To investigate the role of VAMP8 in the vascular transcriptomic response to AngII infusion, we profiled gene expression in suprarenal aortae of VAMP8-knockout (Vamp8-/-) and wild-type C57BL/6 mice receiving 5-day continuous subcutaneous infusion of angiotensin II (AngII, 1,000 ng/kg/min) or saline via osmotic minipump. Four experimental groups were generated: WT+Saline, WT+AngII, V8KO+Saline, and V8KO+AngII (n=4 per group). Differential expression analysis identified transcriptomic changes associated with AngII treatment, VAMP8 deficiency, and their interaction in the aortic wall.

Male littermates were received continuous administration of AngII for 7 days at 0.9ug/hour. Project description:Analysis of angiotensin II effect on left ventricle at gene expression level. The hypothesis tested in the present study was that angiotensin II treatment may affect gene expression in left ventricle in a strain specific manner. Results provide important information about which genes respond to angiotensin II in C57Bl/6N male mice compared to their C57Bl/6J counterparts. Total RNA obtained from isolated left ventricles from C57Bl/6J and C57Bl/6N mice subjected to 48 hours of angiotensin II infusion, via osmotic mini-pumps (500ng/kg/h) implanted sub-cutaneously, compared to sham operated controls.

Project description:Transcriptional profiling of suprarenal aorta from ApoE-/- mice (12-14 weeks old, C57BL/6J background) treated by subcutaneous pump with angiotensin II or saline for 7d, 14d and 28d. Includes Ang II-treated samples at 7d found to have dissected aneurysms. Goal was to examine gene expression in developing AAA in this model over time. Experiment Overall Design: Two condition experiment, one suprarenal aorta per array. Saline vs. angiotensin II at 3 time points, with inclusion of 3 Ang II-treated dissected. Total 35 arrays: 6 saline 7d, 6 saline 14d, 5 saline 28d, 4 Ang II 7d, 5 Ang II 14d, 6 Ang II 28d, 3 Ang II-dissected 7d.

RNA-seq analysis of mouse aortic endothelial cells of angiotensin II hypertensive mice

Project description:To explore the role of endothelial cells in hypertension, we developed an angiotensin II-induced mouse hypertension model. Eight-week-old C57BL/6 mice were anesthetized using 2% pentobarbital sodium, and osmotic pumps were subcutaneously implanted to deliver AngII at a constant rate of 1000 ng/kg/min over 14 consecutive days. Sham-operated mice underwent the identical procedure, with osmotic pumps containing 0.9% saline implanted subcutaneously instead. Subsequently, mouse aortic endothelial cells were isolated and subjected to RNA sequencing analysis.

Project description:Agonist-induced cardiac hypertrophy in TG1306/1R transgenic mice with cardiac angiotensin II overproduction leads to a gradual transition from a compensatory hypertrophic state to heart failure. To gain insight into the molecular mechanisms that play a role in maintaining cardiac integrity in the diseased heart, we performed a comparative study of gene expression between wild-type (WT) and transgenic (TG) hearts using microarray analysis. TG1306/1R transgenics were separated into two phenotypic groups (hypertrophic and dilated) based on morphological parameters (cardiac weight index and histology) and either a moderate (hypertrophic) or a high (dilated) upregulation of molecular markers for hypertrophy and failure, and compared to age and sex-matched wild-types. In this series, twelve 60 week old male TG1306/1R mice were separated into 3 groups: WT1-4= Four Wild-types TG1306/1R littermates genetically negative for the transgene (-/-) Hyp1-4= Four TG1306/1R mice heterozygote for the transgene (-/+) and developing a concentric hypertrophic phenotype* Dil1-4 = Four TG1306/1R mice heterozygote for the transgene (-/+) and developing a dilated cardiac phenotype* This series contains 4 biological replicates for the following triangulation: WT is compared to Hyp, WT is compared to Dil, and Hyp is compared to Dil. * For further information read: Domenighetti AA, Wang Q, Egger M, Richards SM, Pedrazzini T, Delbridge LM.
