Project description:The genetic background of mice used in this study is mainly C57BL/6J with a small contribution from 129/Sv and DBA/2J strains. Male littermates were received continuous administration of AngII for 7 days at 0.9ug/hour. The gut microbiota plays a central role to modulate the plasma metabolome in response to chronic Angiotensin II infusion (part-II)

Project description:Six week old C57BL/6 conventional (mice with gut microbiota; conv, n=6) and germ-free (mice without gut microbiota; GF, n=6) mice were infused with angiotensin-II (AngII) for 4 weeks (400ng.kg-1.min-1; Alzet 1004). In parallel control groups, conv (n=6) and GF (n=6) mice received saline via minipumps. Our primary goal was to identify metabolites which were differentially regulated in conventional mice treated with AngII, but not in GF mice, indicating that these metabolites are microbial in origin. Following minipump implantation, animals were housed singly to prevent cross-contamination of microbiota. At the end of fourth week, feces and blood were collected. Both plasma and feces samples were processed and analyzed by using Liquid Chromatography-Tandem Mass Spectroscopy (LC-MS/MS) for metabolite detection (Metabolon).

Brief Hydrocortisone Infusion Differentially Influences Blood Leukocyte and Monocyte Gene Expression Before and After Endotoxin Administration in Humans (supported by NIH grant GM34695)

Project description:Fourteen human subjects (18-40 years of age) were studied. The control group (saline + LPS [n = 7]) received a 12 hour infusion of saline and the experimental group (cortisol + LPS [n = 7]) received a 12 hour infusion of cortisol (3 µg/kg/min). After 6 hours of saline or cortisol infusion, intravenous lipopolysaccharide (2 ng/kg) was administered to all subjects. Blood samples were collected at -24 hours, 0 hours, 6 hours and 24 hours relative to LPS administration at 0 hours. Leukocytes and monocytes were isolated from these blood samples, total RNA extracted, and microarray analyses performed using Affymetrix Focus GeneChips®.

Project description:Agonist-induced cardiac hypertrophy in TG1306/1R transgenic mice with cardiac angiotensin II overproduction leads to a gradual transition from a compensatory hypertrophic state to heart failure. To gain insight into the molecular mechanisms that play a role in maintaining cardiac integrity in the diseased heart, we performed a comparative study of gene expression between wild-type (WT) and transgenic (TG) hearts using microarray analysis. TG1306/1R transgenics were separated into two phenotypic groups (hypertrophic and dilated) based on morphological parameters (cardiac weight index and histology) and either a moderate (hypertrophic) or a high (dilated) upregulation of molecular markers for hypertrophy and failure, and compared to age and sex-matched wild-types. In this series, twelve 60 week old male TG1306/1R mice were separated into 3 groups: WT1-4= Four Wild-types TG1306/1R littermates genetically negative for the transgene (-/-) Hyp1-4= Four TG1306/1R mice heterozygote for the transgene (-/+) and developing a concentric hypertrophic phenotype* Dil1-4 = Four TG1306/1R mice heterozygote for the transgene (-/+) and developing a dilated cardiac phenotype* This series contains 4 biological replicates for the following triangulation: WT is compared to Hyp, WT is compared to Dil, and Hyp is compared to Dil. * For further information read: Domenighetti AA, Wang Q, Egger M, Richards SM, Pedrazzini T, Delbridge LM. Elucidating the effect of a new indoline derivative (AN1284) against Non-alcoholic steatohepatitis

Project description:The aim of the current study was to check whether chronic treatment with AN1284 could reverse steatosis and fibrosis in a mouse model of NASH. We used a mouse model of dietary induced NASH that was given for 4 month. Treatment with saline or AN1284 was given via implanted minipumps for 2 month. Treatment was started after 2 month feeding. Our results revealed that AN1284 significantly attenuated liver damage, as indicated by a reduced liver/body ratio, decreased ALT serum levels, a significant reduction in liver fat content and hepatic fibrosis. To investigate the underlying mechanism, we performed RNA sequencing on mice fed with normal diet (ND) or high fat diet (HFD; Envigo-Teklad TD.150235).
