Characterization of cholesterol loading effect on the gene expression of bone marrow derived macrophages of AKR and DBA/2 ApoE-deficient mice

Project description:Strain effects on the response to cholesterol loading in bone marrow derived macrophages (BMM) are not well studied. The atherosclerosis susceptible strain, DBA/2 on the ApoE deficient background and the atherosclerosis resistant AKR strains show differences in cholesterol handling when loaded with AcLDL. To understand the molecular basis for these differences, BMM from both strains were loaded with acetylated low desnisty lipoprotein (AcLDL) and gene expression quantified. BMMs from AKR greatly upregulated transcripts involved in the lysosomal trafficking pathway. This experiment strongly suggest that upregualtion of the lysosomal pathway in response to AcLDL maybe protective against atherosclerosis. In addition we find that transcripts containing the sterol response element (SRE) motif had strong strain-loading interactions suggesting a strain response to SRE response.

We already find that some LncRNA can effect cholesterol metabolism and level of inflammation factor, which may influence atherosclerosis process.

Itaconate alleviates cholesterol burden via ABCA1 stabilization and cholesterol efflux

Project description:Background and aims: Itaconate (ITA) is a metabolite produced from the tricarboxylic acid cycle (TCA) that has been shown to regulate atherosclerotic plaque growth and induce stability via immunomodulation. However, lipid metabolism regulation by ITA is currently underexplored in atherosclerosis. Here, we take advantage of plaque-targeting ITA-conjugated nanoparticles (ITA-LNPs) to investigate the effects of ITA on regulating lipid metabolism in foam cells/macrophages in atherosclerosis via ABCA1 stabilization and increased triglyceride metabolism. Methods: Apoe−/− mice were fed a high-cholesterol/high-fat diet (HCHFD) for 12 weeks and injected once weekly with 50 mg/kg ITA-LNP or Ctrl-LNP. Aortas were tested for ITA-LNP biodistribution, followed by quantification of atherosclerotic plaque burden. Bone marrow-derived macrophages (BMDMs) or Raw 264.7 cells were treated with ITA-LNP or Ctrl-LNP in the presence of oxLDL, acLDL, or free cholesterol to investigate ITA’s actions on lipid metabolism and ABCA1 expression under a variety of conditions, including stable gene knockdown. Results: ABCA1 was significantly upregulated with ITA-LNP treatment compared to Ctrl-LNP both in vivo and in vitro at the protein level, but not at the transcriptional level. ITA-LNPs were shown to prevent ABCA1 decay via the HO-1-calpain axis, resulting in significantly increased cholesterol efflux in macrophages. This was further confirmed in Raw 264.7 cells with a stable HO-1 knockdown. Additionally, ITA decreased lipid burden in conjunction with increased expression of Slc25a1 in ITA-LNP-treated BMDMs, suggesting enhanced fatty acid-derived citrate shuttling and increased fatty acid metabolism. Conclusions: ITA-LNPs regulate lipid metabolism in atherosclerosis by inducing triglyceride catabolism and cholesterol efflux.

Analysis of mixed-oxysterol treatment on the THP-1 macrophage

Project description:THP-1 monocytes were differentiated, via PMA treatment, to macrophages. THP-1 macrophages were then treated with a mixed-oxysterol treatment (7-ketocholesterol and 7beta-hydroxycholesterol) for 24 hours. The mixed-oxysterol treatment was mixed to a ratio that represents the cytotoxic oxysterol content of human atherosclerotic plaque. Control and treated macrophages were analysed using both 2-DE with peptide mass-fingerprinting and tandem-mass spectrometry.

Transcriptomics of THP-1 macrophages treated with or without IL-26 for 24h

Project description:Purpose: The aim of this study was to obtain a comprehensive transcriptomic analysis of IL-26-exposed macrophages. Methods: THP-1 derived macrophage were generated by incubating THP-1 cells with PMA (50 ng/ml) for 24h.Then THP-1 macrophages were treated with IL-26 for 24h, which were collected and applied to RNA-sequencing. The constructed sequencing library was sequenced using an Illumina Novaseq 6000 system. The expression of transcripts was calculated as fragments per kilobase of exon model per million mapped reads (FPKM). Results: In total, we identified 1303 differentially expressed protein-coding genes between untreated and IL-26-treated macrophages, including 667 up-regulated and 636 down-regulated genes.
