Effect of tocopherol on caco-2 cells.

ABSTRACT: Caco-2 cells (clone TC7) were cultured for 15 days post confluence on plastic dishes to reach the differentiation, then cells were treated for 24h with alpha or gamma tocopherol enriched mixed micelles, or mixed micelles alone (as control) (Landrier et al Free Radical Research 2008). Cells were rinced with PBS before RNA extraction with Trizol, according the standard protocol of the manufacturer. RNA were checked for integrity with bioanalyser and quantified with Nanodrop. All others steps (labeling, hydridisation, wash and scan) were perfomed with Agilent kits, scanner and dedicated software.

Project description:The aim of this study was to detect miRNAs modified by biactive dietary lipids in a cellular model of enterocytes. The selected model was Caco-2 cells Differentiated Caco-2 cells were incubated with micelles containing the lipids for 24 and 48 hours. The expression leveles of the miRNAs were compared with cells treated with empty micelles. Microarray analysis of HK-2 cells treated with cisplatin

Project description:Chemotherapeutic use of cisplatin is limited by its severe side effects. In this study, we demonstrated that cisplatin induces cell death in a proximal tubular cell line by suppressing glycolysis- and tricarboxylic acid (TCA)/mitochondria-related genes. HK-2 cells were cultured to confluence in 100mm dishes. Total RNA was extracted (QIAGEN, Valencia, CA, USA), and the concentration in the samples was measured using a Micro UV-Vis fluorescence spectrophotometer (Malcom, Tokyo, JAPAN). Sample of 10ɥg of Total RNA from HK-2 cells were labeled with biotin (3'IVT Labeling Kit, Affymetrix, USA) and hybridized (GeneAtlas Hybridization, Wash, and Stain Kit for 3' IVT Arrays, Affymetrix). The Microarray platform used was Affymetrix HG U219 Array Strip. The arrays were scanned in a GeneAtlas scanner controlled by GeneAtlas Software (Affymetrix, Sta. Clara, USA). Genes were later filtered according to expression fold change (Affymetrix Expression Console software, Affymetrix)

Project description:Peripheral blood mononuclear cells were collected from the patients. RNA-seq and analysis was performed on the collected samples. Total RNA was extracted from the tissue using TRIzol® Reagent according the manufacturer’s instructions(Invitrogen) and genomic DNA was removed using DNase I (TaKara). Using bioinformatic analysis, we aimed to identify changes in genes and pathways in human DPSCs and BMMSCs . Cells (BMMSCs and DPSCs) from healthy male subjects were seeded in 10 cm2 dishes and cultured until they reached 80 % confluence, and total RNA of DPSCs and BMMSCs were extracted and analyzed.

Project description:Tocopherols (Vitamin E) are lipophilic antioxidants that are synthesized by all plants and are particularly abundant in seeds. Two tocopherol deficient mutant loci were used to examine how tocopherol deficiency impacts global gene expression during the critical peroid of germination and early seedling development when tocopherols are essential. vte1 lacks all tocopherols, but accumulates the tocopherol pathway intermediate DMPBQ,. vte2 which lacks all tocopherols and pathway intermediates. We used microarrays to examine the global gene expression in early seedlings and identify distinct classes of genes whose expression is affected by tocopherol deficient mutants vte1 and vte2. Project description:To investigate the global effects of vitamin E supplementation on heart aging, we used high-density oligonucleotide arrays to measure transcriptional alterations in 30-month-old B6C3F1 mice supplemented with Î±- and Î³-tocopherol since middle age (15 months). Experiment Overall Design: Gene expression profiles were obtained from 5-month-old controls, 30-month-old controls and 30-month-old mice supplemented with Î±-tocopherol (1g/kg), or a mixture of Î±- and Î³-tocopherol (500mg/kg of each tocopherol).
