Expression data from FACS-isolated lung endothelial cells

ABSTRACT: To investigate the role of a TIE1-function blocking antibody on gene expression of lung endothelial cells. Lung endothelial cells were FACS-isolated from primary tumor-bearing mice. Mice were treated either with IgG or TIE1-blocking antibody during primary tumor growth. Total RNA were used for microarray analysis using Clariom S mouse assays (Applied Biosystems).

Project description:LRG1 is a novel proangiogenic and profibrotic molecule that is upregulated in pathologies characterised by abnormal angiogenesis such as AMD and cancer. Blocking LRG1 by genetic models or by our in-house developed function blocking antibody leads to vascular normalsation. In this study, we have treated B16F0 melanoma tumours with 15C4, our function blocking antibody, or with IgG as a control. The tumours were collected at day 17 and the endothelial cell population was isolated by FACS sorting the CD31 positive cells. RNA was isolated and used for the preparation of cDNA libraries. Expression profile of the endothelial cell population was assessed by Illumina sequencing. Differenital expression analysis was conducted using the DESeq2 package in R.

Project description:To investigate what kind of gene expression is regulated downstream of Tie1 signaling, we performed RNA-seq analyses on venous and lymphatic endothelial cells (ECs) between homozygous tie1 mutant embryos and their wild-type and heterozygous siblings during secondary sprouting, respectively. Our analyses indicate that Tie1 signaling controls a series of gene expression involved in lymphatic development including EC migration, proliferation, and differentiation.
