Project description:Altered levels of circulating extracellular miRNA in plasma and serum have shown promise as non-invasive biomarkers of disease. However, unlike the assessment of cellular miRNA levels for which there are accepted housekeeping genes, analogous reference controls for normalization of circulating miRNA are lacking. The objective of this study was to devise an approach to both identify and validate circulating miRNA reference controls on a de novo basis, and evaluate the relative performance of these customized internal reference controls versus other strategies. As a case example, 1066 different miRNAs were screened in plasma from pulmonary arterial hypertension patients and control subjects to identify an initial pool of miRNA candidates. Circulating miRNome of Trachemys scripta after elective gonadectomy

Project description:Pain management is an important issue in veterinary medicine, requiring biomarkers with high sensitivity and specificity for the timely and effective treatment. Emerging evidence suggests that miRNAs are promising pain-related markers. The aims were to profile the circulating miRNA signature in plasma of turtles (Trachemys scripta) and point out potential candidate biomarkers of pain. Plasma of female turtles underwent surgical gonadectomy were collected 24h pre-surgery, and 2.5h and 36 h post-surgery. The expression of miRNAs was profiled by Next Generation Sequencing and the dysregulated miRNAs were validated using RT-qPCR. The diagnostic value of miRNAs was calculated by ROC curves.

A considerable proportion of hematopoietically-derived miRNAs are present in plasma with the ability to confound the signature of true circulating miRNA species. We use microarray analysis to catalogue a list of 313 haemotopoetic miRNAs and analyze expression profiles of cell-free miRNAs in individual plasma fractions after calibraiting for cellular miRNA signals. Comprehensive global maps of bona fide circulating miRNA species are presented, and inter-individual varibility and gender-specific expession is explored in populations of healthy individuals. Supernatant 1 (S1) fractions from 8 male and 10 female individuals.
