GSK3 activates telomerase by regulating dynamic behaviour of multiple transcription factors converging on the hTERT gene

ABSTRACT: Telomerase controls cell immortality and is a promising cancer target. Understanding transcriptional regulation of the catalytic subunit hTERT is critical to realise effective telomerase therapeutics. GSK3 inhibition suppresses hTERT expression. Overall design: A2780 ovarian adenocarcinoma cells were cultured continuously for 21 days in the presence of DMSO or 2.5mM GSK3 inhibitor 6-bromoindirubin-3’-oxime (BIO) with replenishment weekly on days 1 and 4. Cells were subcultured, counted, and RNA samples taken.

INSTRUMENT(S): Agilent-014850 Whole Human Genome Microarray 4x44K G4112F (Probe Name version)

Telomerase controls telomere homeostasis and cell immortality and is a promising anti-cancer target, but few small molecule telomerase inhibitors have been developed. Reactivated transcription of the catalytic subunit hTERT in cancer cells controls telomerase expression. Better understanding of upstream pathways is critical for effective anti-telomerase therapeutics and may reveal new targets to inhibit hTERT expression. In a focused promoter screen, several GSK3 inhibitors suppressed hTERT repor  ...[more]
