Expression profiling of PaTu8988s and PaTu8988t cell lines

ABSTRACT: We used microarray to look at the genes deregulated in PaTu8988s (adenovirus insensitive) and PaTu8988t (adenovirus sensitive) cell lines PaTu8988s and PaTu8988t are pancreatic cell lines derived from the same patient and show significant responses to oncolytic adenoviruses. The different response to adenovirus in these cell lines is not related to the previously known mechanisms that affect adenovirus infection. The two cell lines represent represent a "suitable" model to screen tumour-associated genes affecting the potency of aoncolytic adenovirus.

Paclitaxel resistance in ovarian cancer increases oncolytic adenovirus efficacy via upregulated CAR expression and dysfunctional cell cycle control

Project description:The microtubule-stabilising drug paclitaxel has activity in relapsed ovarian cancer. However, resistance frequently develops. Oncolytic adenoviruses are a novel cancer therapy, and replicate selectively within and lyse malignant cells, leading to productive infection of neighbouring cells. We found increased efficacy of adenoviruses of multiple subtypes in paclitaxel-resistant ovarian cancer cells. There was increased expression of a key adenovirus receptor, CAR (coxsackie adenovirus receptor), due to increased transcription that resulted from histone modification. Moreover, CAR transcription increased in intraperitoneal xenografts with acquired paclitaxel resistance and in tumours from patients with paclitaxel-resistant ovarian cancer. Finally, we identified dysregulated cell cycle control as a second mechanism of increased adenovirus efficacy in paclitaxel-resistant ovarian cancer and that inhibition of CDK4/6 using PD-0332991 was able both to reverse paclitaxel resistance and reduce adenovirus efficacy. Thus, paclitaxel resistance increases oncolytic adenovirus efficacy via at least two separate mechanisms.

Project description:The aim of our study is to define the role of glycosylation in the interaction of pancreatic tumor cells with dendritic cells and/or in the metastatic behaviour of the tumor cells. We want to examine two cell lines, established from the same primary human pancreatic tumor, which exhibit different grades of differentiation as well as a different potency to metastasize. We believe that these cell lines represent a suitable model for study of glycosylation in the cell biology of human pancreatic adenocarcinoma.
