Glucocorticoids and androgens suppress group 2 innate lymphoid cell activation and protect from pathogenic gastric inflammation and metaplasia

ABSTRACT: We performed scRNAseq of leukocytes isolated from the gastric corpus of WT male mice. CD45+ leukocytes were sorted from the gastric corpus using flow cytometry and libraries were prepared using the 10x genomic chromium controller. These data provide an atlas of the inflammatory cells which reside within the gastric corpus during steady-state conditions.

YAP-activated metaplasia underlies gastric adenocarcinoma and corpus regenerative mechanisms [RNA-Seq]

Project description:YAP is a transcriptional co-activator of the hippo signaling pathway and is known for its oncogenic and regenerative activity across numerous tissue types. In particular, high YAP levels in patients with gastric cancer (GC) confer a lower survival rate and poor prognosis for these individuals. Therefore, there is a great need to develop targeted therapies against these aggressive tumors. However, the role of YAP and its underlying molecular mechanisms during gastric tumorigenesis are still poorly understood. Using genetic models, we demonstrate the oncogenic function of YAP in CLU+ gastric cells in vivo. YAP over-expression in CLU+ cells induced atrophy, metaplasia and hyperproliferation in the gastric corpus, while its deletion in a Notch activated gastric tumor model rescued metaplasia. Furthermore, we defined the YAP1 targetome in YAP activated gastric tumors, and showed that YAP1 binds to the active chromatin elements of spasmolytic polypeptide-expressing metaplasia (SPEM) related genes and activates their expressions in gastric tumors and ulcers. Together, these results reveal YAP1 as a critical regulator of metaplasia in the gastric corpus, and highlights YAP signaling as a possible therapeutic target to inhibit the progression of gastric tumors.

Berberine for the intestinal metaplasia after endoscopic therapy for early-stage gastric cancer

Project description:Interventions: After the study registry, intestinal metaplasia score of gastric corpus upper gastrointestinal endoscopy and gastric biopsy (updated Sydney system) is evaluated, and only cases with 2 or more score continue to the intervention of (1) or (2) by randomized method. The ratio of (1) and (2) is 2:1 with minimization method. In the cases with 1 or less score, the case finish the study. 10x single cell gene expression and V(D)J immune profiling of T cells from the blood, synovial fluid and synovial tissue in psoriatic arthritis patients

Project description:This study shows the pronounced expansion of CD8 T cell clones at sites of active inflammation within the joints of psoriatic arthritis (PsA) patients compared to blood, and characterises the gene expression of expanded clones using droplet based single cell RNA sequencing. Mononuclear cells were separated from freshly acquired paired peripheral blood and synovial fluid samples from 3 PsA patients using density gradient separation, then enriched for CD4 and CD8 T cells using FACS. Additionally, CD45+ leukocytes were enriched by FACS from the cryopreserved synovial tissue of 2 further PsA patients. All cells were then processed using a 10x Chromium Controller and sequenced by Illumina HiSeq 4000 (peripheral blood / synovial fluid) or NovaSeq with S2 flowcell (synovial tissue).
