Microarray expression profile of circular RNAs in cervical cancer

ABSTRACT: Human immortal cervical cell line H8, human cervical cancer cell line SiHa and SiHa transfected with Wnt11-overexpression vector were included in the study. We identified circRNAs differentially expressed between H8 and SiHa or between SiHa and SiHa transfected with OV-Wnt11.

Project description:Five human cervical cancer cell lines (HeLa, CaSki, SiHa, C-33A, SW756) and one human normal cervical epithelial cell line HcerEpic were included in the study. Microarray based circRNA expression profiles were acquired using the Arraystar Human circRNA Array (8x15K, Arraystar). We identified circRNAs differentially expressed in human cervical cancer cell lines compared to human normal cervical epithelial HcerEpic cells (control).

Effect of BRCA1 overexpression on genetic expressions of cervical cancer based on next generation sequencing

Project description:The cells were cultured in DMEM medium supplemented with 10% fetal bovine serum (Sangon, China) and 1% antibiotic-antimitotic solution (Gibco, USA). The SiHa cells were transfected using Lipofectamine 3000 reagent following the manufacturer’s protocol and incubated for 48h for further analysis.The gene expression profiles were analyzed of SiHa cells transfected with GV230-BRCA1 or GV230 plasmid to assess effect of BRCA1 overexpression on genetic expressions of cervical cancer.

Circular RNA profile of human cervical squamous cell carcinoma

Project description:Circular RNAs (circRNAs) represent a widespread class of non-coding RNAs, which drew little attention in the past. Recently, limited data showed their promising future to act as biomarkers in human cancer, but the characteristics and functions remain largely unknown in human cervical cancer. In this study, we demonstrated the expression profile of circRNAs in cervical squamous cell carcinoma(SCC) patients with circRNA microarray, and identified a large number of circRNAs possibly expressed in SCC.

MicroRNA analysis in HPV-positive and HPV-negative cervical cancer cell lines

Project description:Persistent infection by high-risk human papillomaviruses (HPVs) is associated with the development of cervical cancer and a subset of anogenital and head and neck squamous cell carcinomas. Abnormal expression of cellular microRNAs (miRNAs) plays an important role in the development of cancer, including HPV-related tumors. MiRNA expression profile was investigated by microrray analysis in the HPV-positive cervical cancer cell lines SiHa (HPV16-positive cell line derived from a cervical squamous cell carcinoma), CaSki (HPV16-positive cell line derived from a metastatic cervical epidermoid carcinoma), and HeLa (HPV18-positive cell line derived from a cervical adenocarcinoma) and compared with primary HFKs and C33a (HPV-negative cervical cell line).

MicroRNA‑130a‑3p inhibition suppresses cervical cancer cell progression

Project description:MicroRNAs (miRNAs or miRs) play essential roles in the initiation and progression of human tumors, including cervical cancer. However, the mechanisms underlying their actions in cervical cancer remain unclear. The present study aimed to evaluate the functional role of miR‑130a‑3p in cervical cancer. Cervical cancer cells were transfected with a miRNA inhibitor (anti‑miR‑130a‑3p) and a negative control. Adhesion‑independent cell proliferation, migration and invasion were evaluated. The findings presented herein demonstrated that miR‑130a‑3p was overexpressed in HeLa, SiHa, CaSki, C‑4I and HCB‑514 cervical cancer cells. The inhibition of miR‑130a‑3p significantly reduced the proliferation, migration and invasion of cervical cancer cells. The canonical delta‑like Notch1 ligand (DLL1) was identified as a possible direct target of miR‑103a‑3p. The DLL1 gene was further found to be significantly downregulated in cervical cancer tissues. On the whole, the present study demonstrates that miR‑130a‑3p contributes to the proliferation, migration and invasion of cervical cancer cells. Therefore, miR‑130a‑3p may be used as a biomarker to determine cervical cancer progression.

Genome-wide effect of Galectin-7 ectopic expression in HeLa and SiHa cervical cancer cell lines.

Project description:The Galectin-7 gene has been found subregulated in several cervical cancer cell lines and in clinical samples. To study the changes in the cell system after the ectopic expression of Galectin-7, we used microarrays to analyze transcriptome of HeLa and SiHa cancer cell line. Cells were transfected with a viral vector with the Galectin-7 gene and negative controls in 3 biological replicates and analyzed to study gene expression changes. Epigenetic treatment of cervical carcinoma cell lines

Project description:Four cervical carcinoma cell lines (C4I, CaSki, HeLa, SiHa) were treated with epigenetic modifying drugs 5-aza-2’-deoxycytidine and trichostatin A. Such treatment leads to reactivation of genes that are epigenetically silenced in cancer by aberrant methylation; reactivated genes were then identified by microarray profiling. Subsets consist of 2 biological repeats and 2 dye-flip experiments for each repeat for CaSki cell line, and 3 repeats, each with 2 dye-flip experiments for C4I, HeLa, Caski cell lines. The identification of reactivated genes, their selection for validation experiments, and final identification of 6 genes that are significantly more often hypermethylated in cervical carcinoma clinical specimens is described in (manuscript submitted).
